Soybean genetic transformation is very difficult because of the low plant regeneration rate. An effective regeneration system with high-quailty soybean of Hebei was established. The effects of different genetypes, inducing times and the concentration of hormones on explants of embryonic tip of Jidou7, Jidou16, Jidou17 and Jihuang13 were studied, the differences between two regeneration systems, cotyledonay nodes and embryonic tips, were compared either. The results indicated that the ratio of bud and regeneration were decreasing with the increasing concentration of 6-BA, otherwise, there was an opposite tendency about the number of shooting explants. According to our result, the regeneration rate of embryonic tips explants was obviously higher than that of cotyledonary nodes when the concentration of 6-BA was from 0.5mg/L to 1.0mg/L, and the optimal inducing time was 5d. Comparaed to the regeneration system of soybean cotyledonay nodes, embryonic tips has the advantages of higher regeneration rate and repeatability, simple and convenient operation, shorter test cycle and more sensitive to antibiotic.Based on the regeneration system of embryonic tips and Agrobacterium, the transformation conditions with GUS gene were studied. The results indicated that the best condition if OD value of Agrobacterium was 0.6~0.8 and 200umol/L AS was added to the medium in dark cultivante. We analysised the Agrobacterium-mediated transformation Jidou16 by PCR, and the results showed that the objective E2 gene was transformated successfully, transformation efficiency was 5.88%.
