A polyclonal anlibody(pAb)specific for sulfanilamide (SD) was prepared. Hapten SD was coupled with carrier protein human sernm albumin (BSA) to form an antigen SD-BSA by diazotization method. The conjugates used to immunize New Zealand white rabbits were identified with UV scanning spectrum and the couple rate of the antigen was32:1.After the antisera were obtained and ourified and purified using a nodified n-caprylic acid-saturated ammonium sulfate method,the specificity of pAb was detected by indirect ELISA.It showed that the titer of pAb was1:105 and the lowest detectable concentration in terms of 70% inhibition rate was 0.09μg/ml .the standard calibration curve for SD was linear in a range of 1 ng/ml~100μg/ml.the cross-reaction rate between pAb and SD analogs was low. The results showed that the anti-SD pAb had a high specificity and was good enough for immunoassay of SD in edible animal products.
