With the progress in modem Genetics and technique of Molecular Biology, AG gene had been cloned from Arabidopsis thaliana, from then on large-scale studies have carded out on homeotic genes in floral organ development. And then the ABC model was established by researching the homeotic mutation of Arabidopsis thatiana and Antirrhinum Majus. The A, B, C represent three classes hometic genes respectively, take different functions, and possess position effects during the development of flower. Later, the D gene was obtained from Arabidopsis thaliana and Antirrhinum Majus, which is related to the form of placenta and the determination of ovule in the center meristem. As the result, the ABC model was extended to be the ABCD model. There are two A class genes: AP1 and AP2, the B class genes include AP3 and PI, the C class gene is AG. and D class gene is AGL11.However, there are more questions are waiting for being solved, and one of these is how the expression changes in the visible development of buds and blooming. In order to measure the change of expression in buds development, blooming and withered flower, we cloned the partial fragments of ABCD genes for semi-quantitative RT-PCR. This work would be very helpful to demonstrate the change of the ABCD genes at these special stages; moreover it would contribute to the breeding of rape, the creation of male sterile line and the alternation the shape of flowe.Expression analysis of ABCD genes, which related to the development of floral organ in rape (Brassica Napus), was carried out by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR), using rapeβ-Actin gene as a control. Here we obtained the fragments of ABCD genes and built an optimized semi-quantitative RT-PCR system.The result was showed by the IOD ratios of ABCD genes toβ-Actin. The IOD ratios of ABCD genes toβ-Actin exhibited a variation in the expression level of ABCD genes at the different development stages. At the early stage of buds development when the diameter is from less than 1mm to 3 mm, the expression of the six genes all showed an increased trend. But at the next stages, every gene has its particular expression. AP1 expressed the most when the diameter of buds exceed to 3 mm and the length of them are more than 5 mm, the fourth stage. Later then, it appeared gradually decrease in the followed stages. The expression of AP2 arrived at the top at the third stage, and then slowly reduced until the flowers wither. AP3 presented increased expression from the least buds to blooming flower, whereas it showed distinct decrease as the flower withers. PI went up before the petals come out, the following expression also fell. AG reached the top in the third stage and kept stable expressing until blooming. And in the withered flower it showed similar. The appear of AGL11 was the same as AG.
