There are many chemical constituents in the discarded tobacco leaves.And they are active constituents,i.e. solanesol,nicotine,sclareol,tobacco polysaccharide, VE, Vk2. So it has important significance to strengthen the utilization of the tobacco leaves and discarded tobacco leaves.This paper aimed on the comprehensive utilization of discarded tobacco leaves,and mainly engaged in the extraction technology,isolation and determination of active constituents,i.e.solanesol,nicotine,from the leaves of discarded tobacco.The works are summarized five aspects as follows:1 Select methanol solvent for extraction. The orthogonal design of four factors and three levels were used to select the optimal extraction method based on the single factor experiments. The best extraction process is: temperature is 50°C; the extraction times are three;the ratio of material to liquid is 1 '. 6;the extraction time is 3h.The method is stable and easy to operate.The yields of solanesol is 85% in this method.2 A new purification process of extracted solanesol has been developed in this work.Mainly steps are contained in this procedure,i.e.saponification with KOH, silica gel column chromatography and recrystalization.The optimal condition was determined.The results showed that the content of solanesol was high to 96%.3 Optimized the operational condition of TLC of solanesol and sclareol.By studying on the developer,the optimal thin layer condition was determined.It's rapid and reproducible.4 Determine solanesol by RP-HPLC,by selecting the ultra-violet detector, with methanol :ethanol =1 : 1(V/V) as mobile phase,UV examination wave-length is 213nm.The result has shown that it has good linear relationship in the range of 2.08~41.6μg·mL-1(r=0.9999).5 The process of extraction and purification has been developed by studying on other active constituents,i.e.sclareol,nicotine and tobacco polysaccharide.The qualitative and quantitative analysis were set up for sclareol, nicotine and tobacco polysaccharide applied with TLC, GC, HPLC and the method of phenol- sulfuric acid.
