This study was focused on the isolation, purification, structure and physiological activity of polysaccharides from Agrocybe aegerita ( As-1 ) . Three methods—compound extraction, cold-alkali extraction and hot-water extraction—were used to extract polysaccharides from As-1. The result shows that it was effectively useful to obtain polysaccharides from As-1 hyphostroma with hot water, for the cell wall of the hyphostroma is not very compact and the connection between polysaccharides and protein is looser than that in carposporoph. The yield of polysaccharides from As-1 hyphostroma and purity coefficient of polysaccharides extracted were 11.52% and 95.04% respectively. The cold-alkali extraction was best in extracting polysaccharides from carposporophyte, for their tight cell wall and the compact linkage between polysaccharides and protein, but there was much protein contained in the extract. Through the test of hydroxy radical clearance, the result shows that the polysaccharides extracted from As-1 stipe using cold alkali was effectively in clearing hydroxy radical, and the clearance rate was 72.7%.DEAE-cellulose chromatographic method was used to purify the polysaccharides. The result shows that there were 5 sorts of polysaccharides in the extraction from carposporophyte hot water, Z1 was presented large amount, and S1 was found to be the main part in polysaccharides from hyphostroma with hot water. According to FTIR study, the result shows that S1 is sort ofβ-glucan. The glycosidic linkage turned out to beβ- pyran-glucan.In the animal test, it was found that the content of MOD in the mice which were injected with polysaccharides extracted from hyphostroma was striking decreased, and the TAOC, the enzyme activity of SOD and GHS-PX were extremely high, when compared with that of control group. So to speak, polysaccharides extracted from hyphostroma can effectively enhance the ability of anti-oxydation in the organism. But the polysaccharides extracted from carposporophyte showed a poor ability in enhance the TAOC and the enzyme activity of SOD, but it can effectively enhanced the enzyme activity of GHS-PX, decreased the amount of MDA in the liver of the mice, and raised the spleen index significantly. Meanwhile both of these two kinds of polysaccharides could decrease the cholesterin in the serum of the mice, besides they both could boosted the expression of serumβ- andγ-globulins in the serum of the mice.
