The research is based on orientating of phyllosphere dominant microbe using an integrated approach of PCR-DGGE and traditional methods,furthermore studying the character of strain and colony,the physiology and biochemistry characters of strain,the enzymology character,et al,and probed into the parameters of fermented technology of the dominant microbe.Through orthogonal design,set up different artificial fermentation conditions,synthetically analyzed several important biochemical indicators and enzymatic activity indicators,combined with the smoking quality,screened the best fermentation condition.The results summarized as follows:1.The result of the character of strain and colony,the physiology and biochemistry characters of strain,the enzymology character as follows:the character of strain and colony were that the thalli is bacilli form,has motility,can form endogenetic spore, sporangium ill defined enlarged,spore is ellipse and grow in the middle,gram straining is masculine.The colony of the dominant microbe is milk white and opaque, the surface of the colony is fold,and the brim of colony is neat.The important biochemistry characters of the strain as:Anaerobic growth;V.P positive response;No lecithin enzyme;Resistance faintly to lysozyme;Weak salt tolerance;can use sugar or alcohol fermentation acid;degradable starch;to the use of citrate;Weakly reducing nitrate;hydrolysis casein and tyrosine;can not benzene phenylalanine deaminase or less.Concerning the character of producing the enzyme of the nature strain,the findings showed that,the strain has ability to producing catalase,amylase,proteinase, pectinase and so on.Which has provided the rationale foundation in decreasing the starch,the protein and the pectin,improving leaf tobacco quality and taste.2.With regard to fermentation process parameters of the superiority strain,the findings showed that,its growth output is the biggest when the temperature is 40℃and the pH is 8.0.For the nature fungus's growth,the best carbonaceous matter is corn flour,nitrogenous matter is the yeast cream and metallic ion is Mg2+, respectively.3.Main three factors(A,preparation;B,temperature;C,humidity)were studied of influencing the leaf tobacco fermentation quality via the orthogonal table L9(33), some factors were test have the important contribution to the leaf tobacco quality such as chemical targets,the enzyme activity,the smoking quality and so on,the results showed:The best fermentation condition is A3B2C2,namely exerts the superiority fungus and the mix enzyme,the temperature is 35℃,the humidity is 70%.Under this condition,the carbohydrate compounds and the nitrogen compounds reached to the balance,be propitious to the total phenol class matter and the pigment class matter transformation.Several kind of enzyme activities to be at the best conditions.
