| Five species of animal (pig, goat, dog, rabbit and chicken)which representate polyphagia,ruminant, sarcophagy, gnawer and bird species respectively were used as experimental subjects. All ofthe male animals are 1/2 sex-maturity. The method of perfusing ligated intestinal segment in vivo andin situ was conducted to investigate the residual, adsorption and absorption characteristics of soybeanagglutinin (SBA) in the small intestines and the effects on intestinal histological structure in differentSpecies of animal with different SBA concentrations.Purified SBA was isolated through the linked guar-amylose, which was disposed to Krebs liquidwith different SBA concentrations. Using the method of perfusing ligated intestinal segment in vivoand in situ to infuse the Krebs liquid to the jejunum of animals which were began with 1/3 relativeposition to the anterior of jejunum (pig, 130cm; goat, 130cm; dog, 40cm; chicken, 10cm;rabbit,30cm). There are four sections with the length of 5cm each in one unit,and four units in onespecie. Meantime, the SBA was set up with four concentration gradient: 0.0mg/mL,0.5mg/mL, 1.0mg/mL,3.0mg/mL, also four rePeats. The perfusate volume was disposed withthe stand of intestinal surface area.(pig, goat and dog,SmL; rabbit and chicken, 2mL). After 8 hours,the indirect inhibiting ELISA method of soybean was used to detect three measure indexes: theresidual amount of SBA in perfusate, the surface adsorption amount of SBA in intestinal mucosa,the ingress absorption amount of SBA in intestine. In order to compare the absorption extent in allspecies of animal, the percentage was adopted to react the result.The results showed that from the analysis of residual rate, adsorption rate, and absorption rate,the survey indexes diversed significantly(P<0.05) in one kind of animal with differentconcentrations, and also diversing significantly (P<0.05)in the same concentration with variousspecies. The residual rate of SBA in all kinds of animals increased with the rising of SBAconcentration, and reached to the max in 3mg/mL(P<0.01). The lagest adsorption intensity is in3mg/mL also. Rabiit, chicken, goat had the largest absorption rate in 1mg/mL. All kinds of animalshad the largest absorption ability in 0.5mg/mL accept from rabbit, which was in 1mg/mL. As for theadsorption ability, pig and dog got to the max in 1mg/mL, as well the others were in 0.5mg/mL. Thisindicated that the sensitivity of SBA to animal's intestines was stronger at low concentration, the adsorption ability and absorption ability were comparatively large. Among the three concentrations,goat had the largest residual rate, the lower adsorption rate and the lowest absorption rate. Rabbit hadthe largest absorption rate, lower residual rate and adsorption rate. So it showed that the permeabilityof SBA to rabbit's intestine was higher. Chicken had the same tendency, but the permeability wasworse than rabbit's, its residual rate and adsorption rate got to max in 3mg/mL. Pig and dog has thelargest adsorption rate among the animals, it showed that the adsorption ability of SBA to intestinalmucosa membrane was comparatively large. As for the respect of SBA concentration, we can see thatlow concentration play a more important role in the effect to intestine. All the kinds of animals hadthe better adsorption effect and absorption effect in low concentration(0.5mg/mL 1mg/mL), whenSBA concentration reach to 1mg/mL, rabbit, chicken, and goat had got to the max in absorptionintensity, and reduced in 3mg/mL. And pig and dog could continue to absorb in 3mg/mL, but itsextent is little.From the histological section result, the effects of SBA on jejunum in various animals were notobvious. It is possible to relate to the short experimental session. All the same, it could indicate thatwhen adding SBA to the jejunum, villous height decreased and crypt, depth increased. Because thesurvivability of SBA diverse among kinds of animals, the concentration of SBA to the effect alsodifferent. |
