On the basis of F2 population of hybrids between common buckwheat (F. esculentum) variety Sobano and F. esculentum var. homotropicum accession, genetic markers on RAPD, STS and some morphological relative traits were studied and their genetic linkage maps were constructed. The main results were as follows:1. The map of F. esculentum var. homotropicum has eight linkage groups with 54 DNA markers and 3 morphological markers, covering 526.6 cM. The map of common buckwheat Sobano (F. esculentum) has eight linkage groups with 48 DNA markers and 2 morphological markers, covering 443.1 cM.2. Homo-style, shattering habit, and acute achene traits are all controlled by the single dominant gene Ho, Sht, and Ac respectively, integrated successfully to the genetic linkage maps. Among them, Ho and Sht genes are linked on the linkage Group 1, and Ac gene on the linkage Group 4.3. There are some RAPD Markers close to morphological markers. For example, S64-601 is tightly linked to Ho gene and S52-1106 tightly to Ac gene, which are useful on buckwheat breeding.4. The RAPD and STS marker fingerprint maps of common buckwheat Sobano and F. esculentum var. homotropicum accession were constructed, which are helpful to variety or parent identification. |
