The Duck Swollen Head Septicemia Virus (DSHSV) is one member of the family reoviridae, genus Orthoreovirus, the average size of the isolated virus is 65~80 nm. It is a dsRNA virus without envelope. It's dsRNA is over 19kb length. In 2002, This virus was the first reported in China. The main symptom of diseased ducks were swollen head and diffuse hemorrhage.This experiment selected age of duck embryo, concentration of trypsin and concentration of FBS in DMEM. Through repeated experimenting, we apply 14-day-old duck embryos, trypsinization with 0.125% trypsin and 15%FBS in DMEM to culture Duck Embryo Liver Cell (DELC), 20-day-old duck embryos, trypsinization with 0.25% trypsin and 20% FBS in DMEM to culture Duck Embryo kidney Cell.This virus is no cytopathic effect (CPE) in Duck Embryo Fibroblast(DEF) and Duck Embryo Kidney Cell (DEKC) for five passages, The cytopathic effect characteristics of DSHSV in Duck Embryo Liver Cells is begin to appear after 24h at the fist time, half of cells fall off from the culture flask after 48h. The propagation of DSHSV was stable in Duck Embryo Liver Cells (DELC) by five passages. The cytopathic effect characteristics of DSHSV in Duck Embryo Liver Cells showed the particle increasing in cells, cellular swelling, and fall off from the culture flask.The rarefied DSHSV mix with Freund's Adjuvant to prepare hyperimmune serum.We can observe that the virions were nonenveloped spheroidal particle and the average size of the isolated vires was 65~70 nm. In addition, it was assessed back to duck embryo, we find diffuse hemorrhage in the duck embryos and duck's bill severe distortion. This pathological changes is the same to else reports, those Experiments shows that CPE in Duck Embryo Liver Cells is caused by the duck swollen head septicemia virus.
