Mastitis is one of the most common diseases in diary cow. It effect not only the production ofmilk to cut down the economic returns but also the lactation ability of diary cows. Some sick cowshave to be killed at last. Sometimes it does harm to the quality of milk and human health.Diary goat was taken as model in this study. In this study change of lymphocyte andmammary epithelial cells apoptosis in mastitis of diary goat and other different deal groups wasobserved, and induced the subclinical mastitis model with Staphylococcus aureus. Dependablity oflymphocyte apoptosis in peripheral blood with time after intramammary instillation ofStaphylococcus aureus was detected. Pathogenesis of mastitis was observed from the way ofapoptosis, to provide the new way and theory for precaution and therapy of mastitis.In this study flow cytometry was used to detect the change of lymphocyte apoptosis inperipheral blood in mastiffs of diary goat group and other deal groups. The results showed that thelymphocyte apoptosis rate (%) in peripheral blood of mastitis group was significantly higher thanother groups (P<0.01, P<0.05), vaccine group was significantly lower than mastitis group (P<0.01), Chinese herbal medicine group was significantly lower than control group (P<0.01, P<0.05), Combination group was significantly lower than vaccine group (P<0.01, P<0.05) and wasnot significantly different with Chinese herbal medicine group (P>0.05).In this study flow cytometry was used to detect the change of Splenic lymphocyte apoptosisin mastiffs of diary goat group and other deal groups. The results showed that the lymphocyteapoptosis rate (%) of Splenic in mastitis group significantly higher than other groups (P<0.01, P<0.05), vaccine group was significantly lower than mastitis group (P<0.01), Chinese herbalmedicine group was significantly lower than control group (P<0.01, P<0.05), Combination groupwas significantly lower than vaccine group (P<0.01, P<0.05) and was not significantly differentwith that of Chinese herbal medicine group (P>0.05).In this study flow cytometry was used to detect the change of lymphocyte apoptosis rate ofLymphonodi supramammarici in mastiffs of diary goat group and other deal groups. The resultsshowed that the lymphocyte apoptosis rate (%) of iymphonodi supramammarici in mastitis groupsignificantly higher than other groups (P<0.01), vaccine group was significantly lower thanmastiffs group (P<0.01), Chinese herbal medicine group was significantly lower than controlgroup (P<0.01, P<0.05), Combination group was significantly lower than vaccine group (P<0.01,P<0.05) and was not significantly different with Chinese herbal medicine group (P>0.05). In this study flow cytometry was used to detect the change of lymphocyte apoptosis rate oflymphonodi mesenterici in mastitis of diary goat group and other deal groups. The results showedthat the lymphocyte apoptosis rate (%) of lymphonodi mesenterici in mastitis group wassignificantly higher than other groups (P<0.01), vaccine group was significantly lower thanmastitis group (P<0.01, P<0.05), Chinese herbal medicine group was significantly lower thancontrol group (P<0.01, P<0.05), Combination group was significantly lower than vaccine group(P<0.01, P<0.05) and was not significantly different with Chinese herbal medicine group (P>0.05).In this study TUNELwas used to detect the change of apoptosis of mammary epithelial cellsin mastitis of diary goat group and other deal groups. The results showed that the apoptosis ofmammary epithelial cells in mastitis group significantly higher than other groups (P<0.01),vaccine group was significantly lower than mastiffs group (P<0.01, P<0.05), Chinese herbalmedicine group was significantly lower than control group (P<0.01, P<0.05), Combination groupwas significantly lower than vaccine group (P<0.01, P<0.05) and was not significantly differentwith that of Chinese herbal medicine group (P>0.05).In this study subclinical mastitis model was induced by Staphylococcus aureus, 1~36h afterintramammary instillation, lymphocyte apoptosis rate (%) in peripheral blood increasedsignificantly, 36~96h a little lower, after that it kept stable.
