Nowadays the tomato cultivation area extends continuously and diversificationof planting mode, it caused many illness in tomato.Tomato Leaf mold is one of the most seriously diseases, specialty in the protective(greenhouse and plastic house). it is also a worldwide disease, which has caused heavy losses intomato production. if chooseing and identification resistance breeding by people it needs muchtime and nicety is not high. so it will nicety and speedingess using molecular markers for theresistance gene in tomato leaf mould Cf-19 gene. and it will help the next works of resistancbreeding and gene cloned, basic for cultivating resistance breeds.The Cf-19 gene of resistance tomato leaf mould is found in this years, nowadays research areless about it. in this paper using AFLP molecular marker to marker the Cf-19 gene,expect findingclosely linked to gene Cf-19. and using this linked marker to find resistance germplasm in thetomato germplasm resourse for prepared for resistance breeding, the result of research are follows:1. HN19 is resistance materials and HN16 is not resistance materials, HN19×HN16 F2population materials. he resistance pattern of a F2 population derived from cross of HN19 andHN16 was analyed thrugh inoculation with Cladosporium fulvum.The response patterns showedthat R:S segregation ration for 3:1,Suggesting that the resistance gene was a single dominant gene2. 545 AFLP primers combinations were screened and PCR-amplified products were isolatedon and through BSA screening finding 25 AFLP primers combinations. using these primercombinations to using HN19×HN16 F2 progenies to develop marker s for Cf-19. Five AFLPmarkers(E38M61-2,E66M84-4,E66M44-1,E77M 50-1,E77M50-2) closely linked to gene Cf-19were acquired. E38M61-2 was found closely linked to the Cf-19 locus at a distance of 9.7 cM<10cM.3.using AFLP marke to screen the tomato germplasms,at the same time identifying theresistance in the greenhouse.There are 15 of 95 tomato germplasm resoures.using AFLP marker 3Of 15 are resistance.This the AFLP marker improved is credibilty. This result of AFLP markers isbase for molecular breeding and fuction of gene cloning. |
