| Mx1 gene was found in a inbed mouse strain A2G is resistant to influenza virus. Theinterferon -induced Mx protein has broad-spectrum antivirus ability. It is one of the best studieddeterminants of innate immunity to viral infection. In the infection of the influenza virus, it canbind to NP protein in order to thereby inhibiting influenza virus multiplication. So it have antiviralactivity against influenza virus. Now Mx1 was used of a candidate gene for disease resistance.Swine influenza(SI) is a important disease to pig industry. The pathogen of SI is Swineinfluenza virus (SIV). It can reside in pig which have infect with SIV or mucago of air passage. Inour country, The scale level of pig industry is low, when the pig infect with SIV, it can not outbreak.So the pig have lower antiviral activity against influenza virus. But with level higher than before, itmay outbreak Nowadays, we must take more attention to antiviral activity breeding.In this experiment we have cultivated swine airway epithelial cell, SIV can multiplicat in it. Aproper system was established for induced Mx1 with interferon. Then, we detect the expression ofthe Mx1 use Real Time PCR. We find the proper time and the level of the interferon. And we alsodetect the early expression of Mx1 in cell which infect with SIV.The results are as follows:(1) the system of DMEM/F12 medium+10%FCS+1%antibiotic is proper for cell. And theSIV can multiplicat in cell.(2) The expression of Mx1 is increas with the increasing level of the interferon, induced Andthe most proper level is 1000IU/ml.(3) The expression of Mx1 is not increase with the time of the interferon induced. It reach thehighest level in 4h. Then down slowly.(4) The early expression of Mx1 in cell which infect with SIV also is not increase with thetime of the interferon induced. It reach the highest level in 4-6h. Then down slowly. And thetendency also similar to the process of human. It demonstrate that, human and swine may infectedwith same disease.
