| 'Nubiana' plum (Prunus salicina Lindl.) was used as the materials in the study. Through analysis of the optimal multiplication medium combination, the factor on rooting; sterile system was established using the shoot -tips and the stems as explants.We studied plant growth regulators and their combination, basal medium, and different carbons which affected on the callus inducing 'Nubiana' leaves in vitro, and studied the regeneration from petiole primarily. The main results were as follows:Firstly, through the analysis of the explants' rate of survival and the times of multiplication, the optimal medium was LP+6-BA 0.4 mg/L+IBA 0.08 mg/L;and through analysis of the effect of Ad concentrations on explants multiplication and growth ,the optimal concentration of Ad was 1.5~2.0 mg/L.Secondly ,through analysis of the effect of the IBA concentration, different basal medium and carbons on shoot rooting, the best rooting medium was 1/2 WPM+IBA 0.5 mg/L+ glucose 15 g/L. It was better to place them in darkness for 7 d before transfering to white light.Thirdly, the effects plant growth regulators and their combination, basal medium, different carbons and other factors on callus induction of 'Nubiana' leaves were studied in order to establish the optimum condition of inducing and growth of callus. The results showed that: WPM + BA 0.5 mg/L + 2,4-D 2.0 mg/L+ glucose 30~50 g/L was the best medium for the inducing and subculture of callus;0.075~0.1% citric acid had remarkable results of inducing callus, the inductivity reached 100%, 0.1%CA showed the best result in controlling the browning callus, compared with other browning-resistant materials.At last, adventitious bud was regenerated from petiole of 'Nubiana' for the first time, and the regeneration frequency was 3.3 % on WPM+6-BA 2.0 mg/L+ NAA 0.5 mg/L... |
