In this study, we took the morphology investigation as the foundation, the geneticdiversity of 18 cultivars and 8 R. coreanus varieties were analyzed. The results were asfollows:1. Three methods for DNA extracting of Rubus were compared, including SDSmethod, CTAB method and nuclear DNA method. We found nuclear DNA method was thebest one, because the DNA yield and purity was very high, the DNA color was white. TheDNA was qualified for RAPD amplification.2. The suitable reaction system and amplification procedure were found. PCRreactions were carried out in 25μl volumes containing per reaction. 20ng template DNA,1×buffer, 2.0mmol·L-1 Mg2+, 0.6μmol·L-1 primer, 0.24mmol·L-1 dNTPs, TaqDNApolymerase 1.5U. PCR reactions were carried out under the following programmeconditions:45 cycles at 94℃for 4min, 94℃for 1min (predenaturation), 36℃for 1min,and 72℃for 2min, followed by one cycle at 72℃for 5min. Amplification products wereresolved on a 1.5%agarose gel run in 0.5×TBE buffer.3. 20 primers which amplified 251 repeatable loci (including 235 polymorphic loci)were screened out of 85 primers. The percentage of polymorphic bands was 93.6%. Thelong of fragments were between 200bp and 4000bp, mostly between 400bp and 2000bp.The RAPD fingerprints from 20 primers can identify all cultivars and varieties exactly.4. According to RAPD markers, genetic distance was computed. All materials can bedivided into 4 groups which were in consistence with that of the analysis of morphology.Division of subsets in some groups were related to the geography difference, fruit colorand reproductive characteristic of Rubus.5. In this study, reproducibility and stability of RAPD was proved. The phylogeny ofRubus can be analysised by more random primer loci, which was good for recensing andauthenticating morphologic classification.
