??Tomato was one kind of vegetable which was grown all over the world, its delicious flavor,rich nutrition, fresh color and medical value attracted more and more consumers. With the enlargedtomato cultivating area and improper planting mode, tomato vertucullium wilt have become themain disease which threatening tomato's production. At present, managing vertucullium wiltdisease depends mainly on chemical controlling in tomato production. This method was usually notvery effective and also could cause environment pollution, and can't controlling this diseasesentirely. Therefore, breeding resistant cultivars and improving the resistance of tomato couldcontrol the vertucullium wilt effectively. Many studies have been done about tomato vertuculliumwill abroad, but less studies on molecular biology have been done in china.In the experiment, molecular marker technique was applied to investigate resistant genes ofvertucullium wilt in tomato, screening AFLP and SSR molecule marker linked to vertucullium wiltresistant gene, establishing the AFLP and SSR molecule marker technique system of tomato, andthe marker was used to screen tomato plants resistant to vertucullium wilt.The results establishedthe basis for MAS and cloning of the gene,and then the tomato materials which contained resistantgene could be screened out.The main results of this study is as following:1.In this study tomato resistant cultivar 05046 and susceptible cultivar 051355 were used andtheir hybridizing F2 progenies were used as the separating groups. Using M. incognita as inoculationmaterial, and using conventional inoculation method to establish resistance and susceptibility genepools.2.F2 population gained from 05046x051355 were used to construct the genetic linkage mapusing SSR and AFLP molecular markers. Four new DNA markers linked to the vertucullium wiltresistant gene were identified in the study. Three AFLP markers E66M84-A,E78M84-D andE66M40-A were 10.3cM,14.2 cM and 30.5 cM apart from V. Wilt resistant gene differently.OneSSR markers SSR599 was 12.5 cM away from V. Wilt resistant gene.3.The AFLP molecule markers E66M84-A was used to screen 95 tomato germplasm,meanwhile artificial inoculate identification was executed to identify resistant and susceptibleplants.The inosculation rate is up to 90.5%. The results validated the practicability of AFLP markerand it established the method of molecule marker assisted selection.
