| Mycoplasmal pneumoniae of swine(MPS), caused by Mycoplasma hyopneumoniae (Mhp), has been described in pigs all geographic areas. The disease is characterized by high morbidity and low mortality rates in intensive swine populations systems. During the past few decades, it has been a primary factor associated with pneumoniae being the most economically important disease in finishing pigs. Antibiotic and vaccine are effective to the disease, but corresponding more attention pay to the safe of food and reinforcing of bacteria drug resistant, antibiotc is no longer a wisdom choice. As to vaccine, it is not permissive to use antibiotic within two weeks after intrapuaahmmally inculated the domestic attenuated vaccine, that restricts its application and extention. While the inward inactivated vaccine, the culturists can not afford to it bacauseof its high price. As we know, scientist is focusing their sight on on genetic engineeringvaccine, since the titre of mycoplasma hyopneumoniae culture is low, and needed concertration before making vaccine, which is very troublesome, however there is no any commercial vaccine of mycoplasma hyopneumoniae in the world.Based on the nanometered of prolis and optimition culture of mycoplasma hyopneumoniae,this experment firstly study on the prolis inactivated vaccine of mycoplasma hyopneumoniae.The whole experment is consisting of two parts. Mycoplasma hyopneuminiae were isolated in Frris medium from infected pigs of a hogpen in Shandong Province, which suspected to be Swine enzootic pneumoniae, and identified by PCR test and biochemical test. The correlation of CCU and PCR count of passaged culture is also studied. The results suggest that this isolated strain is mycoplasma hyopneumoniae; and the homologous analysis of P46 gene and P65 gene indicated that the isolated strain has highly homology with J strain,232 strain and 7448 strain, which is 98.47 %, 98.37 %,98.07 % and 99.17 %, 98.95 %, 99.11 %, and corresponding homology of amino acid is 98.36 %,98.36 %,98.03 % and 98.67 %, 98.83 %, 98.83 %.The minimal dilution that PCR can detected is 10-3, which is lower four titre than CCU, when the Mhp culture's CCU is 107. Flollowing the isolated strain is submited to Shandong Binzhou Institute of Animal Science and Veterinary Medicin, which make inactivaed vaccine with it after optimization culture and concentration. Three batch vaccine is used to study the safy and validity.In addition, it's effectiveness is compared with home-made inactived vaccine and inward inactived vaccine. The result is show us that psychosis and diet is normal, there is no swelling and tissue necrosis in the inoculated locus, and no negtive influence on the weighting (P>0.05) and breeding. The propolis vaccine can more promptly peomote immunological response than other two vaccniae in the experiments. In the seventh day after inoculated, the average anbody level of propolis vaccine is higher than other two kinds vaccine(P<0.05), but the the difference is slight(P>0.05) in the next 56 days. The result of lymphocyte transformation assay suggests that propolis can commendably stimulate animal's celler immunity. The stimulative index of propolis vaccine is higher than domestic vaccine(P<0.05), and slight higher than inward vaccine (P>0.05). This experimental result preliminarily tells us that propolis vaccine is likely to instead of inward vaccine. |
