Salviae Miltiorrhiza Bge. was a kind of crucial medical herb, considering the secondary metabolites in it, such as tanshinone and salvianolic acid, which could be effective to treat cardiovascular diseases. Yet mostly secular Salviae Miltiorrhiza Bge.were wild and harvest was blindfold and unplanned, resulting in breads being farraginous and germplasm resources being destroyed badly. So it was cried for studying, exploiting and protecting the germplasm resources.Ten populations of Salviae Miltiorrhiza Bge. from different areas were selected in the research, optimizing Salviae miltiorrhiza DNA extraction methods, RAPD-PCR reaction system and amplification procedure, then several special primers were screened to construct the DNA fingerprintings which could be regarded as an identification criterion to distinguish Salviae miltiorrhiza and other plants and different populations of Salviae Miltiorrhiza Bge. from different areas.The genomic DNA amplification results with the method of RAPD-PCR were analyzed by data statistics and clustering analysis, and the genetic relationships among different populations of Salviae Miltiorrhiza Bge. were obtained; the clustering result of genomic distance was more similar with the real genetic relationship than the similarity coefficient; and the populations could be divided into three main branches in genomic distance. The contents of tanshinone IIA and salvianolic acid B were determined by HPLC, the tanshinone IIA contents of populations in every branch were analogical in trueborn areas, and tanshinone IIA contents were higher compared with non-trueborn areas. The results indicted that synthesizing tanshinone IIA was decided by inheritance for the different populations in their trueborn areas; moreover it was the results that the plant evolved to adapt environments and were selected by nature for a long time, and the results must consider the growing state of plant as well as the supply of carbohydrate. So the adaptability to circumstance of Salviae Miltiorrhiza Bge. could be estimated by tanshinone IIA content difference in trueborn areas and non-trueborn areas. The Salviae Miltiorrhiza Bge. populations from Bo-zhou of An-hui province and Beijing acclimated to the introduction environment according to the smaller difference of tanshinone IIA compared with trueborn areas in the research.Some problems, which should be continued to study, were advanced. Firstly, it was needed to further prove the regulation that it synthesizing tanshinone IIA was decided by inheritance and influenced by growing state and the supply of carbohydrate, The content of tanshinone may reach the highest in the best suitable areas (trueborn areas); on the other hand the content could decrease in unsuitable areas. Secondly, it was essential to research the relationship between environmental factors and quantity of tanshinone IIA and salvianolic acid B and the mechanism how the main environmental factors effect the synthesization of tanshinone and salvianolic acid.
