Triticum durum Desf.has 28 chromosomes, with 42 chromosomes triticum aestivum is the different variety,its output abdicates to the triticum aestivum,the sown area approximately composes the world wheat total area 10%,is only inferior to the triticum aestivum world second big kind of wheat crop. Its protein content compared to triticum aestivum high 1~2 percentage point,the wet gluten content is higher than triticum aestivum ,specially richly contains the kind of carotenoid,has the nutrition and the health care function.The triticum durum or the wheat quality improvement important idioplasm resources,the domestic and foreign wheat breeding the protein,the wet gluten content and the kind of carotenoid content takes the triticum durum improvement of varieties the important target,Therefore this experiment mainly carries on the research analysis from the triticum durum carotenoid and the protein two aspects,thus provides the theory basis for the triticum durum quality breeding. The main results were as follow:1.In order to enhance in the solution the kind of carotenoid in triticum durum to raise the effect,put into cellulose and pectinase in solution to withdraw the condition the optimized experiment;Uses the different solvent,time,temperature,times as well as liquid-solid compared to carry on experiment,and optimal best withdraws the parameter.The generalized analysis and considered the cost element, the carotenoid withdraws the most superior conditions:By acetone: Benzine=2:1 ,the temperature 45℃,each time withdraws 1.5h,expects the liquid-solid compared to 1:10, and joins cellulase and pectolase in turn in the solution,soaks raises 2 times,may obtain best withdraws the effect.2. This experiment through ultraviolet obviously spectrophotometric method and highly effective liquid phase chromatography analysis research the carotenoid.Ultraviolet obviously spectrophotometric method quantitative analysis the carotenoid,by characteristic absorption spectrum determination the carotenoid biggest determination wave length in 445nm position.And carries on the quantitative analysis to the domestic and foreign 24 triticum durum idioplasm resources carotenoid,the obtained test result carries on the difference multiple comparison with SAS software Duncan.but the HPLC was used to quantitative and qualitative analyse a kind of carotenoid in triticum durum with a symmetry300TM (3.5um,4.6x100mm)column which he temperature was 25℃,The mobile phase was the mixed solution of acetonitrile-methanol-n-hexane (70:20:10)with the flow rate of 1.0ml/min, Determination wavelength was 445nm and the sample injected volume was 10uL;So provided that a simple and accurate method to determine carotenoid in triticum durum,can make each component of the durum wheat,carotenoid get the good separation.3. The research triticum durum different growth time of carotenoid usually dynamic accumulation rule,carries on the determination from the grain grouting time-the premilky ripe period–the milky ripe later period -ripeness stage-mature stage,finally indicated:Is gradually is decreasing progressively from the grouting time to the mature period carotenoid,this has the close relation with the kind of carotenoid photosynthesis performance.Its photosynthesis mechanism will carry on in the later research further explores.4. The NIRS was determined the triticum durum protein in triticum durum protein research,the starch and the gluten content in under the powder essence grain premise,has fast realized fast,the simple on-line analysis,is the quality analysis first choice method.The result indicated that,The triticum durum crude protein content average in 14.71% above,in which is bigger than the mean value 14.71% materials to account for the total 66.7%,the starch mean value is 67.45%,the grain wet gluten mean value is about 29.33%;This and on the international triticum durum protein,the starch and the wet gluten standard is consistent. 5. Using half seed wheat HMW subunits to result in the SDS-PAGE analysis method,take Chinese sping,Xiaoyan NO.6,AimengniuI,Zhongyou16 as the comparison,carries on the appraisal analysis to 38 triticum durum's HMW subunits,finally demonstrated:On the Glu-A1 position spot,the triticum durum HMW subunits variation type take null primarily,null as the superiority subunit variation type;On the Glu-B1 position spot,the triticum durum HMW subunits variation type appears the frequency take 6+8 subunit as to be highest,reaches 36.82%.7+8 subunit is next, the frequency is 34.21%,but 13+16, 14+15 these two kind of subunit in the triticum durum for the rare subunit,appears the frequency comparatively to be low (respectively is15.79% and 13.15%);13+16 very is specially rare on the triticum aestivum,because the triticum durum does not include D genome ,therefore does not have D genome encoded 5+10,2+12 and so on the subunit;This test result showed that,the triticum durum and triticum aestivum's HMW subunits has its obvious difference,in particular on Glu-B1 position spot.
