Aphis gossypii Glover( Homoptera:Aphididae ) is an important kind of Agricultural pest, which distribute widely and have wide hosts in the world, can cause badly losses of cotton, melon and fruit, each year. Its wide harm is nearly related with polarization of species. Aphids have many polarizations only in body color, such as the yellow, the green and the intermediate color etc in cotton Aphids. Based on the cotton Aphids body-color, many important conclusions have been obtained as a result of much work done from ecology, genetics and molecule-biology etal by many national and international scholars except that the protein level and the correlation between the body color types and the heat shock proteins which hasn't been reported recently. Meanwhile, it was studied that it isn't instantaneous to change with the change of temperatures by Hui-yan Zhao, but to complete slowly through many generations. How can it survive through overcoming stressing temperature, whether it is correlative with antioxidative enzymes and HSPs or not, which is still unknown.In order to reveal the relationship between the changing colors and the HSPs, and the effects on cotton Aphids in different body-color, useing the two types of cotton Aphids( the Yellow and the Green ) as laboratorial materials, stressing them respectively with different changing temperatures in manual climate-boxes. Analysing effects between the Yellow and the Green in the areas below: Biological parameters, forbearing character, total protein contents and HSPs, respectively. The results are as following:1. After heat stressing, no significant differences were observed for net reproduction rate, with the condition becoming more furious, the length of a generation is longer, the intrinsic rate of increase is smaller, compared with the control at the same body color type. The Yellow and the Green have the biggest length of a generation which is 7.256 and 8.192d, and have the smallest intrinsic rate of increase which is 0.3581 and 0.3426, respectively, at 35oC for 1d. The net reproduction rate and the length of a generation of the Yellow is 13.059±0.56 and 7.0765±0.22, which is significantly lower than those of the Green is 16.135±0.44 and 7.8525±0.35, but the intrinsic rate of increase is no significant differences between the two body color types. There exists a significant difference between the survival rate and the stress temperature and time to the two types. After stressing, the survival rates of both the Yellow and the Green are the highest, are 98.72% at 15 oC for 1d and 97.48% at 30oC for 1d, respectively. Within the same stressing condition, there is no significant difference between them. There is no significant difference in their intrinsic rate of increase and survival rate which shows there are no differences between the two body color types in genetic power.2. A significant difference of the total protein contents of the treatments and the control was observed: showing the highest value between the Yellow and the Green is 109.99μg.mg-1 at 10oC for 30min and is 108.46μg.mg-1 at 30oC for 30min respectively. There is no significant difference between the two color types at the same stressing condition.3. There is an obvious difference on the activities of antioxidative enzymes, compared with the control. With the stressing temperature more furious, their activities are stronger. With the treating time longer, the activities of SOD and CAT are stronger, and it is the most increased rate from 30min to 1h, POD tended to increase at beginning and to slow finally. Under the same stressing condition, it shows no obvious difference between the two types.4. Analysing heat shock protein shows: The Yellow is short of a protein band on 35KD, which exists in the bands of the Green, shows genic slience. Meanwhile, it isn't affected for a protein band on 41.1KD between the two body-color by stressing temperature. With the increasing stress, the bands of normal proteins became thinner and lighter while ones of the Hsp60 turned wider and deeper. But there is no significant difference between them. At the same stressing condition, the stressing temperature affects more greatly on the Yellow than the Green.The cotton Aphids body-color change is the same gene for different expression. The Aphids of body-color change attribute reaction norm of gene types in different environments is a representation diversity, shows that environment is the condition of body-color representation diversityIn conclusion, the experiment is the first to study on heat shock protein in ecology of cotton Aphids, finding the changing rule about protein in cotton Aphids stressing, establishing the changing model on total protein in special condition, and finding the important result that the body color change doesn't relate with Hsp60, which provides theoretical sense for later human. |
