The histological structure of adenohypophysis during reproduction period andnon-reproduction period in mature mullet and crucian was studied and paraffinsection method and frozen section method was used in this experiment. With regard toparaffin sections, H.E. staining method was used to observe the histologic structure ofadenohypophysis, Azan (Mallory-Heidenhain-Azan) staining method, PbH (plumbumhematoxylin) staining method, PAS-OG (periodic acid schiff's orange G) stainingmethod and Jafri combined staining method for fish pituitary was used to display theendocrine cells in adenohypophysis specially and stereology method was used tomeasure and calculate the percentage, volume density, average volume and numericaldensity of the endocrine cells in adenohypophysis. With regard to frozen sections,histochemistry coloration technology was used to show the alkaline phosphatase(AKPase), acid phosphatase (ACPase) and Mg2+ activated adenosine triphosphatase(Mg2+-ATPase), and morphous analysis software was used to measure and calculatethe gray scale value in adenohypophysis. SPSS13.0 statistics software was used toanalysis the above-mentioned results in statistic. The results indicate that:1) The pituitary is of front-back style in mullet and dorsoventral style in crucian.2) There are no distinct, differences between the same kind of endocrine cells ofadenohypophysis in percentage, volume density, average volume and numericaldensity using different kinds of pituitary staining method during reproduction periodand non-reproduction period in the same kind of fish (P>0.05).3) There are distinct differences between mullet and crucian with the endocrinecells of adenohypophysis in percentage, volume density, average volume andnumerical density during reproduction period and non-reproduction period using thesame kind of pituitary staining method (P<0.05).4) There are no distinct differences between the reproduction period andnon-reproduction period in percentage, volume density, average volume andnumerical density with adrenocorticotrophic hormone (ACTH) secretory cells andprolactin (PRL) secretory cells in rostral pars distalis (RPD), thyroid stimulatinghormone (TSH) secretory cells in proximal pars distalis (PPD) and melanophorestimulating hormone (MSH) secretory cells in pars intermedia (PI) ofadenohypophysis in mullet and crucian(P>0.05); There are distinct difference betweenthe reproduction period and non-reproduction period in percentage, volume density, average volume and numerical density with growth hormone (GH) secretory ceils andgonadotropin hormone (GtH) secretory cells in proximal pars distalis (PPD) ofadenohypophysis in mullet and crucian (P<0.05).5) There are distinct differences with three kinds (AKPase, ACPase andMg2+-ATPase, identity in follows) of enzymatic activity in three corresponding parts(RPD, PPD and PI, identity in follows) of the adenohypophysis in the same kind offish during reproduction period and non-reproduction period (P>0.05); There aredistinct difference between mullet and crucian with three kindsof enzymatic activityin three corresponding parts of the adenohypophysis during reproduction period andnon-reproduction period (P<0.05).6) There are distinct differences between the reproduction period andnon-reproduction period with three kinds of enzymatic activity in three correspondingparts of the adenohypophysis in mullet and crucian (P<0.05).
