In this study, the effects of different length of time of sterilization, the recipe of basic medium, the concentration of exogenous hormone(ie. 6-BA, NAA) and different kinds and doses of nutritional additives (coconut milk, banana juice) on the protocorm-like bodies (PLBs) induction rate and proliferation rate of flower stalk buds, plantlets leaves and root shoot tips of Phalaenopsis aphrodite cultivar GC-209, by using in vitro cultivation and statistical analysis method. A culture system was established to induce flower stalk buds of P. aphrodite. A serial of culture media was screened out, which were suitable for induction and proliferation of P. aphrodite PLBs from different explants. This technique could be valuable to some extent for establishment of in vitro micro-propagation system of P. aphrodite.The results showed that:①an optimal cultural medium that induce flower stalk buds of P. aphrodite vegetative buds was: Hyponex No.1 3g/L+6-BA 5.0mg/L+CM 150ml/L+ Sucrose 15g/L+ Active carbon 0.1g/L, pH5.4;②an optimal cultural medium that induce leaves PLBs was: Hyponex No.1 3g/L+6-BA 5.0mg/L +NAA 0.1mg/L+CM 150ml/L+Sucrose 15g/L+ Active carbon 0.1g/L, pH5.4;③an optimal cultural medium that induce leaves PLBs propagation was: Hyponex No.1 3g/L+6-BA 5.0mg/L the +NAA 0.3mg/L + CM 150ml/L +Sucrose 15g/L+ Active carbon 0.1g/L, pH5.4.④optimal explant fit for PLBs induction was young leaves in vitro.In addition, the effects of mutation induced by 15-120Gy 60Co-γrays radiation on P. aphrodite PLBs was also studied. The effects ofγrays on survival rates, propagation rates, and differentiation rates on P. aphrodite PLBs were also analyzed. A primarily conclusion was made that the half lethal dose (LD50) of 60Co-γrays on P. aphrodite PLBs was about 50-68Gy, which could offer an important reference for radiation breeding of P. aphrodite.
