| 3-hydroxy-3-methylglutaric acid coenzyme A reductase(HMGR)is the crucial rate-limiting enzyme relating to cholesterol metabolic process and cholesterol synthesis. The genome DNA was extracted from the blood of Wanxi-White gooses.A pair of primers was designed based on the conservative regions in exon 5 for forward and exon 6 for reverse according to HMGR gene sequences in human,chicken and pig.The intron 5 of HMGR gene was cloned and sequenced,and SNPs of which were tested by PCR-SSCP. SNPs association with goose important economic traits was analyzed.The results as follows:1.The intron 5 was cloned and sequenced,which the length was 704bp registered in the GeneBank(Acession No.EF595972).2.By the PCR-SSCP,allele A and B were tested in intron 5,and AA and AB genotypes were found,the genotypic frequencies of which were 0.5413 and 0.4569 respectively.The frequency of allele A and B were 0.7716 and 0.2284 respectively.3.SNPs association with goose important economic traits was analyzed.The results indicated that the AA genotype increased significantly chest muscle weight,gland stomach weight(P<0.05),diameter of muscle fiber(P<0.01),1000-downs weight and waterproof capacity of feather-down,and decreased chest muscle 45pH and down hold-water percentage.The effects of AB genotype on decreasing chest muscle weight,gland stomach weight,chest muscle percentage,1000-downs weight(P<0.05)and fiber diameter of chest muscle(P<0.01),and increasing chest muscle 45pH(P<0.01)were significant.Therefore, AA genotype was advantageous in improving feather-down waterproof and development of chest muscle and gland stomach,but AB genotype could improve the meat quality in goose. |
