Light is one of the factors which have important effects on growth of plants and.for Light not only is energy for photosynthesis,and light signal for photomorphogenesis,but also regulates secondary metabolism accumulation.Dendrobium huoshanense is a kind of precious traditional Chinese medicine in Anhui Province,and is protected by government for its medicinal value.In order to optimize condition for cultivation of D.huoshannese,enhance medium composite content and provide the technical support for D.huoshannese plantlet industrialization production,a series of researches were conducted by using D.huoshannese plantlets,which were the effects of subculture time and light on plantlet growth and chemical components.Results were showed as follows:1.Making sure 40 days is the best subculture time for plantlet culture of D.huoshannes.The reason was as follows:The height of plantlet increased 282.86%,the tiller increased 3.5 times,fresh weight reached its maximum,content of plantlet chlorophyll almost reached its maximum after 40 clays subculture;water content and sugar content of medium was decreased to the lowest point,pH value of medium could not be suitable to the growth ofplantlet.2.According the test,the best condition for plantlet growth and chemical components accumulation were 30μmol·m-2·s-1light intensity and 12 hours light time per whole day under red light.3.The yellow light improved the growth of stem of D.huoshannes plantlet but the effects of red light and blue light is contrary.Under the red light,the ratio of chla/b of D.huoshannes was low,which had shading biochemistry tendency,but under blue light, the ratio of chla/b of D.huoshannes was high,which had sunning biochemistry tendency.Under red light and blue light,the polysaccharides content was higher,which showed red light and blue light were advantageous to the polysaccharides accumulation of D.huoshannes.
