The chemical fertilizers and the organic synthesis of pesticides to soil structure, ecological damage and environmental pollution,make people to recognize the awareness of agricultural safety.After our county joins World Trade Organization,the problem of agricultural product pesticide residue is obvious and become the foremost limited factors to china's agricultural exports.Now the tendency that green food acts as a center role in production and consumes,and as chemical substances in agricultural application of more and more restrictions,for the production of agricultural production plays an important role in plant growth regulator has indeed made a higher demand.The international food and agriculture organization listens attentively to humanity's aspiration,and clearly put forward the planting shall not use chemical fertilizers and pesticides,forcing many workers engaged in such research to identify natural microbial and biological pesticide to protect the human environment. The Trichoderma for the metabolism of plant material is not only the disease is a disincentive to promote plant growth,but also to improve the long-term use of chemical fertilizers to the soil structure damage,agricultural pollution and endanger people and livestock conditions,may become chemical growth regulator of the greatest potential alternative one.Trichoderma harzianum is the biocontrol fungus with the broadest application in Trichoderma.In the course of biocontrol application,many investigators found that it can promote plant growth.In this research the wild Trichoderma harzianum spore were mutagenized by injecting N+ ion,and one strain called Trichoderma harzianum H-13 was obtained,which could metabolize plentifully certain substance promoting plant growth.Take solid fermentation of Trichoderma harzianum H-13 study, explore Trichoderma spores efficient fermentation conditions and application; Through the various nutritional substances,the culture of Trichoderma harzianum H-13 product of the growth and impact of these compounds,Screening out cheap solid fermentation medium and optimized fermentation conditions for solid, Preparations for the commercial production of Trichoderma provide a theoretical basis and technological parameters.The optimal solid-state fermentation conditions for biocontrol agents of Harzianum Trichoderma strain H-13 were studied,the influencing factors on the conidial concentration were investigated,including carbon,nitrogen,phosphate,the solid material to water,inoculums and the temperature of fermentation.By single factor and orthogonal experiments,the optimal conditions for solid-state fermentation as following:the proportion of bran to main and solid material to water are 3:1 and 1:0.4(g/mL)respectively,sucrose 1.5%,peptone 1.5%,ammuonium nitrate 1%, potassium dihydrogen phosphate 0.05%,NaH2Po4 -Na2HPo4(0.5%-0.50%), inoculum9%(mL/g),the temperature of fermentation among 22~25℃.Under the fermentation conditions,the spore number is up to 1.0×1010/ g after 9 days. Discussion:compared with liquid fermentation,the solid fermentation produces conidiophores have the characteristic of the manufacture simpler,the less cost,the conidiophores content higher is easier to preserve and transport.Influence of the preparation of Trichoderma harzianum H-13 strain on the physiological and biochemical traits of maize were studied with the maize cultured in potted plant.The result showed that,With application the fermentation of the Trichoderma hazianum H-13 strain,the maize Seeding fresh growing,the adult plant fresh weight and dry weight,the content of chlorophyll,the root system vigor,the activity of CAT,POD and SOD in maize seedling were increased,at the same time, the root system vigor,he activity of CAT,POD and SOD target differences and so one reach the remarkable level(P<0.01).Inclusion:the experiment illuminated the preparation of the Trichoderma harzianum H-13 strain to the maize seed's fresh growing trend,the maize seeding growth period various physiologies and biochemistry target have the remarkable promotion influence.Revealed Trichoderma is of a great potential on promoting plant growth.The optimal ultrasonic extraction technology of gibberellins from Trichoderma solid-state fermentation was studied,the influencing factors on the extraction of gibberellins were investigated,including power of ultrasonic,temperature of the material,action time,Concentration of ethanol,ratio of raw to solution and pH value. By orthogonal experiment,the optimal extraction technology was obtained as follows: power360W,temperature 45℃,action time 35min,Concentration of ethanol is 30%, ratio of raw to solution as1∶20 and pH 2.0.Compared with conventional methods,the ultrasonic extraction technology can avoid the gibberellins decomposition under high temperature and get higher 1.7 fields while consumed less time.Moreover the method of the gibberellins extraction could be widely applied in labs and related research,and has the certain reference value to the actual production.In a word:this research the wild Trichoderma harzianum spore were mutagenized by injecting N+ ion,and one strain called Trichoderma harzianum H-13 was obtained,which could metabolize plentifully certain substance promoting plant growth and Trichoderma harzianum H-13 as a fermentation bacteria,by examining various nutrients,fermentation conditions on the Trichoderma harzianum strain H-13 product growth and the effects of these compounds.Solid fermentation taken to explore ways of Trichoderma spores efficient fermentation process and its application; Screening out cheaper solid fermentation medium and optimization of solid fermentation conditions;Pot trial confirmed Trichoderma agents use dosage form; The optimum extraction of ultrasonic assisted GA process,Trichoderma preparation for the production and commercialization of industrial and agricultural production in the application of theory and technical parameters.
