Genetic diversity of 30 microsatellite were analyzed in 13 indigenous black-bone chicken breeds of china. Allele frequency, effective number of alleles, heterozygosity, polymorphism information content were calculated. Clustering dendrograms were obtained from two genetic distances (DS,DA). The resules were as follows:1 A total of 248 alleles were detected at 30 microsatellite loci in 13 indigenous black-bone chicken breeds of china. Number of alleles ranged from 4 (MCW150) to 12 (ADL136,LEI0094), 8.2667 per locus. Mean effective number of alleles was 4.1403.2 All loci showed high polymorphism, mean PIC was 0.6851. Among 30 microsatellite loci, there were 17 loci higher than 0.7, 27 highly polymorphic loci and 3 mediately polymorphic loci.3 The genetic diversity of each breeds was very rich and was of high selective potency. The average heterozygosity of the lueyang chicken is the highest (0.6954), and that of the Jiuyuan Black chicken is the lowest (0.6410).4 Mean Fst of 30 microsatellite loci was 9.95%, which indicated that 9.95% of the total genetic variation came from breed differences and the rest came from individual differences. Except the locus of MCW0330 was deletion heterozygote, the rest was residual heterozygote. Gene flow of 30 microsatellite loci was 2.2628.5 The results of two genetic distances were completely consistent. The distance between Jinhu Silky and Velutinate Silky was the closest that was 0.1552 (DS) and 0.1459 (DA), the distance between Jinhu Silky and Jiuyuan Black was the farthest that was 0.4419 (DS) and 0.4337 (DA).6 Dendrograms based on DS and DA seemed to be similar. The 13 indigenous black-bone chicken breeds of china were clustered into five groups. The first group included Jinhu Silky, Jiangshan Silky, Velutinate Silky and Xingwen Silky; the second group included Yanjin Silky, Muchuan black Silky and Yunyang white Silky; the third group included Lueyang and Silky; the only difference between the two clustering methods was that Yugan Silky clustered with Jiuyuan Black in the UPGMA tree but clustered with Zhuxiang and Wumeng Silky in the NJ tree.
