Xylocarpus granatum belongs to the genus of Xylocarpus Koenig ,the family of Meliaceae. It's the only one Xylocarpus Koenig plant in our country. Xylocarpus granatum is distributed in the mangrove foreast closed to the tropics sea coast in Hainan province. There are some limonoids in their fruits, which have showed extensive bioactivities such as antifeedant, anticancer and antivirus. The object of this is research mainly to find out new lead compounds with pesticide bioactivity from the leaves of Xylocarpus granatum, providing the lead compounds for the new pesticide formulation. We expect to discover new pesticide through simulation experiment and modified synthesis.The results were showed as follow:1,Comminuting the air dried leaves and immersing 3 times by the ethanol. The extracts are concentrated to the cream . The extract were extracted with petroleum ether, ethyl acetate and n-butanol solvents respectively. Then the extracts were isolated by column chromatography,thin layer chromatography, recrystallization and identified by modern spectrum methods(IR,1HNMR,13CNMR and EI-MS).4 compounds were isolated and identified from the leaves of Xylocarpus granatum . They are triacontanol,β- sitosterol, 3-(1-hydroxyethyl)-4,4- dimethyl-4- butyrolactone and Kaempferol-3-O-β-D- glucoside. All the four compounds were isolated from this plant for the first time, and the 3-(1-hydroxyethyl)-4,4-dimethyl-4- butyrolactone is a novel natural product. The main chemical composition is Kaempferol-3- O-β-D-glucoside.2,The essential oils from the leaves and fruits of Xylocarpus granatum were extracted by steam distillation. The components of the essential oils from the leaves and fruits of Xylocarpus granatum were analyzed by GC-MS techniques, of which 33 and 36 compounds were identified for the first time from the leaves and fruits, resepectively. The main compounds of the essential oils from the leaves wereβ-selinene,γ-selinene,α-edrene,β-farnesene andα-patchoulene and from the fruits wereβ-selinene,γ-selinene, d-cadinene,α-bergamoten andδ-cadinene.The main components of both the essential oils wereβ-selinene andγ-selinene.3,The petroleum ether extract , ethyl acetate extract, 1-butanol extraction of the leaves and 3-(1-hydro-xyethyl)-4,4-dimethyl-4-butyrolactone were put into the PSA culture medium and PDA culture medium to examine their fungistatic activity to the Rhizoctonia, Phytophthora, Botrytis cinerea, Gossypii anthrax and so on. The preliminary bioassay showed that the three extractions have fungistatic activity against the Botrytis cinerea. The ethyl acetate extraction has inhibitory rate of 71% at the concentration of 50mg/L. The new compound of 3-(1-hydro-xyethyl)-4,4- dimethyl-4- butyrolactone has the inhibitory rate of 30% to the Phoma asparag. |
