In this study, one strain of Anaplasma was isolated from naturally infectedgoat in Du'an of Guangxi. The 16SrRNA gene sequence of this isolate wasanalysized by the preparation of genome DNAs, PCR amplification, sequencingand compared to 23 strains of Anaplasma deposited in GenBank to inferred aphylogenetic tree. The A. ovis isolated in Guangxi were separated into an A.ovis cluster with other 6 strains from China, America and South Africa, theclosest sequence was 16S rRNA gene of A. ovis-Idaho AF318945 from America,with a similarity of 100%.The analysis demonstrated that Anaplasmosis oviswas discovered in Guangxi first at molecular level,and this strain was same of A.ovis-Idaho AF318945 from America possibly.Based on the sequence of this research and the published sequence of 16SrRNA gene of Anaplasma, a pair of primers of a PCR diagnostic method weredesigned and synthesized for epidemiological investigation of Anaplasmosis inGuangxi. PCR detection technique was suit to detect earlier or invisible infection of Anaplasmosis demonstrated by the specific test and sensitive test.And the blood sample could be detected store at 4℃six monthes or ordinarytemperature 2 monthes.In the epidemiolos investigation of Anaplasmosis ovis, 121 blood samplescollected from Nanning, Liuzhou, Du'an, Dahua of Guangxi were detected byPCR diagnostic method and the test under micros cope. There are 9 to the byPCR diagnostic method and 20 to the test under microscope gave positive tests.In this test, there are some false-positive or false-negative samples tested undermicroscope. Because this method induced false results was affected byconditions of test or stand of subjective jugement. By the PCR diagnosticmethod, there are 6.3% positive samples from Liuzhou and 20.6% from Du'an,no one from Nanning and Dahua. This result determine that Anaplasmosis oviswas epidemic in some places of Guangxi, and it was serious infected. So someefficacious measures of prevention and cure should be utilized after a extensiveand deep investigation.
