| The phosphatidylethanolamine-binding protein gene from Saussurea involucrata Kar.et belongs to anti-cold revelant gene.Its expression production is revelant to the stability of cellular membrane,and make an important role in the cold resistance of plant.We constructed the plant expression vector according to the strategy of constucting the plant expression vector. XLPEBP gene was included in the vector.We transfer XLPEBP gene into cotton utilizing pollen tube pathway method and Agrobacterium-mediated transformation. The main results were listed as follow:1 construction of the plant expression vector:We constructed the plant expression vector including XLPEBP gene, promoter CaMV35S and GUS gene.2 establishment of a highly efficient regeneration system of cotton:(1)We transfer XLPEBP gene into cotton utilizing Agrobacterium-mediated transformation and study all kinds of factors which affected information.(2)The optimum concentration of kanamycin used in resistant selection.3 examination of transforming regeneration cotton:The results of GUS demonstrated the integration of target gene into cotton genome primarily.4 detection of transformation plant:We extract the genome DNA of resistant plant to amplify by PCR.It has been proved that the integration of XLPEBP into cotton genome.5 transformation of pollen-tube pathway method:Two types cotton were selected as the acceptor genotype. We got transformation plant and harvested many seeds(T1)by a series of examinations. |
