Abstract/Summary:
Laboratory animal science is the foundation and key support of life science. Theoccurrence and development of human diseases are of extremely-complex, therefore, ascalled "the living precision instruments" the laboratory animals are indispensable foraccurate research on the pathogenesis and treatment of human diseases. Rodents, particular,rat, not only is extensively used, but also has its unique advantage in comparative medicine,in establishing animal models for a variety of human diseases, investigating some humandiseases, understanding the mechanism of disease and in searching drugs and therapeuticapproaches. Clean rodents must be used in development of drugs, biological products,pesticides, food additives and teaching according to our state standard. However, thequalified animals need be equipped with matched animal care and laboratory environment,the result of the experiment conducted under such conditions, thus, will be of morescientific, accurate, and reproducible. Therefore, the objective of this study is to evaluatecurrently very extensively used individual ventilated cage system but not yet with statestandard as well as to make scientific judgments in hope to provide scientific base for usingthis system in animal care.TestⅠcomparison of physiology and biochemistry of SD rats in a barrier systemand in Individually ventilated cagesObjective: to compare biological features of SD rats in a barrier system to in theindividually ventilated cages. Methods: The reproduction of 10 pains of SD rats from abarrier system and individually ventilated cages were evaluated, and the blood routinevalues and blood biochemical of 50 days rats from a barrier system and individuallyventilated cages were examined. Results: There is no significant difference in growth development, breeding and reproduction for rats in barrier system and individuallyventilated cages; The weigh gain for male rats in early 30 days after birth in barrier systemis more obvious than that in IVC'S, but not 40 days after birth. There is for no differencefemale after birth 30 days. 2. Significant difference in some blood items is observed forrats under these two conditions including that the levels of PLT,POT,PDW,MPV andLPCR in plasma on the blood routine valves are significantly increased or decrease in ratsin barrier system compared to IVC (P<0.01). The levels of Tbil,ALP,Cre,GLU,Cu2+,Zn2+,Na+, Cl-,CO2 is significantly increased while LDH,CK,CK-MB,LCL-C,APOB,lipase, Mg2+ decreased (P<0.05). Conclusion: There is some difference for growth,development and some the blood items for rats in barrier system and IVC.TestⅡcomparison the main organ weight and coefficients of SD rats in a barriersystem and individually ventilated cagesObjective: To measure and analyze the main organ weight and coefficients of SD ratsin a barrier system and individually ventilated cages. Methods: total 60 SD rats wereused in this study, 30 in each group. The rats were raised in a barrier system and IVC for120 days. The main organ weight and coefficient for heart, liver, spleen, lung, kidney;Adrenal, Brian, Testis uterus, ovary were then measured using an electronic scales.Result: Between female SD rats, spleen weight showed significant difference (P<0.01), allorgan coefficients were no significant differences (P<0.05); Between male SD rats, allorgan weight were no significant difference (P<0.05), kidney coefficient showed significantdifferences (P<0.05); Between female and male, in a barrier systems and IVC, all organweight showed very significant differences (P<0.01), Between female and male in IVC, nodifferences were found in the liver and kidney coefficients (P<0.05); Between female andmale in close system, the organ coefficient of the heart, spleen, lung, adrenal and brainshowed significant the difference (P<0.05). Conclusion: The organ weight andcoefficient of SD rats in a barrier system and IVC were observed no difference. TestⅢAnalysis the micro-environment of rat's IVC-B system in working stateObjective: To study the relationship between temperature, humidity, number ofanimals and ammonia change and to provide the base for scientific animal care. Methods:daily recording changes of temperature, humidity, collecting air samples and analyzingthem according to GB 14925-2001. Result: in yearly temperature between 19℃-24℃, therelative humidity in IVC system is 40%-97%, the level of ammonia increases with thenumber of animals as well as with days. Some goes beyond level, not meet state standard.Conclusions: no more 3 rats (weight between 180-300 g) should be in one cage under IVCsystem, and the bedding should be changed once two days.
