Melampsora larici-populina Kleb. is also called leaf rust and widely distributed in the world poplars planting areas. It is one of the most widely distributed and dangerous diseases. It has harm to Populus, black poplar group, even Populus euphraticao and other various poplar trees. There are obvious differentiation and complex genetic background in this fungus. Five Melampsora larici-populina. races have been reported by Europe and North America, China have also found the phenomena of differentiation in this fungus races. However, previous studies did not find the original disease-specific molecular markers. This thesis mainly from the following aspects to do research makes a breakthrough in the Molecular marker research on Chinese Melampsora larici-populina.Firstly, optimization of RAPD system. As we all known, RAPD has its own drawbacks when it was used to pathogen DNA molecular markers, such instability, vulnerability to environmental conditions. Furthermore, there are obvious differences in the result after different laboratories and laboratory making. These shortcomings caused great inconvenience to the exchange of results of RAPD study, almost not for the. In the principle of optimizing RAPD system and exchanging the RAPD test results easily, by optimized various factors of RAPD reaction system carefully, finally to confirm a more appropriate RAPD reaction system.Secondly, finding differential molecular markers. Chinese scholars have done a lot of research in Melampsora larici-populina. But the research involved specific molecular markers in Melampsora larici-populina is relatively small. The test on the basis of screening a large number of random primers, to screen CMLP1, CMLP3, CMLP4, and CMLP5 race-specific RAPD markers by using Optimal RAPD system.Thirdly, the establishment of CMLP1, CMLP4, and CMLP5 Race SCAR marker. Through a further transformation for specificity of RAPD Marker which had been found to attempt to building Chinese CMLP1, CMLP4. and CMLP5 Race SCAR marker, which have been get successfully after testing. Providing a more effective method for rapid identification, epidemic monitoring and detection for Chinese Melampsora larici-populina.
