Wheat leaf rust is an important disease of wheat, which threats and hinders the wheatproduction. In this research, two-dimensional electrophoresis was established to studythe different proteomic expression between TcLr10 and Thatcher. Then MS was used toanalysis the proteins of different expression. Then we can get the amino acid sequencesof these proteins which can provide guidance to breeding resistant varieties throughgenetic engineering.The resistant near-isogenic line TcLr10 against wheat leaf rust and susceptible parentsThatcher was used in this research. Extracting the proteome of TcLr10 and Thatcherinoculation and with out inoculation of Puccinia triticina. The optimized 2-D systemadapted to wheat was established. TCA-acetone was used to extract the proteome of thesamples. Using acetone to precipitate proteins four times could get the better results. Thetime of the two equilibration steps were all 15 min. Following this, it could reduce theeffect of the vertical streaks.The 2-D maps of TcLr10 and Thatcher inoculation and with out inoculation ofPuccinia triticina were analyzed. The results indicated that there were four sports ofdifferent expression between them. MALDI-TOF-MS was used to analyze the four spotsof proteins, and searched their homologous proteins in the database based on NCBI.These proteins may have function in the resistant responses of wheat to Pucciniatriticina.(1)The protein spot "a" showed remarkable similarity with H~+-transporting ATPase,subunit 1. The enzyme is localized on plasma membrane. H~+-ATPase performs animportant function to produce the H~+ gradient of the membrane. It may play animportant role in signal transduction in the interaction between wheat and Pucciniatriticina.(2)The protein spot "b" remarkable some similarity with RuBisCO large subunit-bindingprotein subunit alpha. It is chloroplast chaperone protein-a molecular chaperones. Itmay perform an important function to protect the functional proteins and play animportant role in signal transduction in the interaction between wheat and Pucciniatriticina.(3)The protein spot "c" showed remarkable similarity with putative phospholipidhydroperoxide glutathione peroxidase. It can specifically make the Phospholipid Hydroperoxide reduction. The enzyme can repair the peroxide damage of the host inthe interaction between wheat and Puccinia triticina.(4)The protein spot "d" showed remarkable similarity with DEAD/DEAH boxhelicase-like. It plays a role in nuclear transcription, pre-mRNA splicing, degradationof RNA. It may initiate the transcription and expression of resistance-related geneswhen the plant is suffering from the stress.
