Study On Technology Of The Extraction And Chemosynthesis Of Hypericin

Hypericin is one of the most important active components of hypericum perforatum .It found that hypericin was of significantly effect of antidepression, inhibition central nerve, antiviral activity, anticancer, Its source is aim at two aspects: abstraction of hypericum perforatum and chemical syntheses. Both of them were reported nowadays, but the former existed complexs of abstraction and separation technic,the latter existed lower field, higher cost, etc., and it made industry of hypericin hampered. So the study composite literature date in and out of country to improve abstraction and chemosynthesis route .Synthetically route has been designed as follows:1. AbstractionAbstraction and separation of hypericin adopted two steps to eliminate miscellaneous object, then which was abstracted by ethyl acetate. abstrction solution was condensed to the extractum,Through colophony volume layer chromatography (VLC), hypericin was respectively separated by 40％and 80％ethano, collected the red section liquid, reduced pressure concentration to obtain hypericin extractum.. In the whole process, we adopt TLC trail experiment, finally hypericin content was determined by HPLC.2. ChemosynthesisThis experiment adopts acetone and ethyl formate as initiation material,via a serial of chemical reactions,such as condensation reaction, methylation reaction, acylation reaction, and reduction reaction to synthesize target compound—hypericin. To identificate structural correctness,we determined chemical constitution of significant intermediums, for instance 2-hydroxy-4-methyl benzoic acid, 2-methoxy-4-methyl benzoic acid, emodin and hypericin, by M.P, IR,MS and ~1HNMR.3. AssayingWe groped repeatedly to establish suitable experiment condition which was adopted to measure hypericin content ,and strictly according to pharmacopoeia(2005) rule. We carded out assaying to hypericin extractum and hypericin synthetics which were extracted and synthetized by ourselves.Results as follows:1. Make use of acetone and ethyl formate as initiation material to synthesize hypericin via a serial of chemical reactions, it's yield was31％and it's content was 85％and greatly cut down cost. At the same time, we got several important chemistry intermedium.2. The hypericin extractum. from hypericum perforatum L. have lower hygroscopicity, lower glutinosity. the yield and content of hypericin were 8.8％and 1.2％respectively and the latter was greatly exceeded the international standard(0.3％).It was concluded that the whole synthetic route and extraction route of hypericin, which were formulated in experiment, were feasible and offer favourable guidance for industry.
