| Conjugated linoleic acid(CLA)is a mixture of geometric and positional isomers of linoleic acid which has many kinds of physiologic function,especially potent repartitioning effects.The previous study in our lab showed that the growth performance was not affected, but abdominalfat rate reduced significantly,while meat quality improved with CLA diets. in yellow broiler chickens.The regulatory mechanism about hyperplasia and hypertrophy of adipose cell is very complicated.The regulators about fat metabolism include growth factors,hormones,transcription factors and enzymes.Therefore,the present study,the effects of CLA on the growth performance,slaughter parameters,serum leptin and adiponectin,GHR,adiponectin,AdipoR1,IGF-1,IGF-1R and PPARγmRNA espression in adipose tissue of Yellow broiler chickens were determined.Our study was aimed to reveal the effects of dietary CLA on fat accumulation of broiler chickens and its molecular mechanism.A total of 60(half male and half female)one-day-old Yellow broiler chickens were allocated to two dietary treatments of 30 chickens each with different inclusion level of CLA(0%and 3%CLA)and were fed for 49 days.The chickens were slaughtered in each group on day 49 respectively.Blood samples were collected from the vena jugularis total and some abdominal fat was put in liquid nitrogen.Slaughter parameters,serum leptin and adiponectin,the expression of GHR,adiponectin,AdipoR1,IGF-1,IGF-I R and PPARγmRNA in adipose tissue were measured.Body absolute weights,average daily gain, average daily feed intake and the ratio of feed to gain of all broilers were determined at the beginning and end of the experiment.The results as fellows:1 Dietary CLA inhibited growth performance of female chickens.Average daily gain was reduced respectively by 6.36%on d 28 and 7.02%on d 49(P<0.05)on CLA diet.2 Dietary CLA increased leg muscle weight.And leg muscle index was increased by 9.37%of male and 15.92%of female respectively.Abdominal fat rate was reduced by 22.05%(P<0.05)female on CLA diet.Other slaughter parameters were unaffected by CLA.3 Dietary CLA increased serum leptin level(P<0.05),but adiponectin level was unaffected.4 The abundance of adiponectin and AdipoR1 mRNA expression in abdominal adipose tissue of chickens was unaffected by CLA. 5 Dietary CLA reduced abundance of GHR mRNA(P<0.05),but it didn't effect abundance of IGF-1 and IGF-I R mRNA in abdominal adipose tissue of chickens.6 Dietary CLA sharply inhibited the expression of PPAR gamma mRNA in abdominal adipose tissue of chickens(P<0.01). |
