| Porcine endogenous retrovirus (PERV) is a member of Mammalian Type CRetroviridae, integrating into the genome of porcines by the form of previrus DNA andreplicating with the replication of cell chromosome. In 1997, Patience's researchconfirmed PERV could infect human-derived cells in vitro. As a result, the safety ofXenotransplantation was on the agenda.So we detected PERV among GuangXi Bama(GXBM) minipig group using themethod which had been established in other domestic laboratory, aimed at selecting andbreeding PERV negative minipig groups, providing safe and reliable organs forxenotransplantation. 114 blood samples were collected from GXBM minipig anddetected by PCR. The results showed that PERV was universal in the minipig and thepositive rate is up to 100ï¼…and didn't find PERV negative individuals. Among them,subtype A and subtype B are count to 91.4ï¼…and 87.7ï¼…respective. None of thembelong to subtype C. The main structure proteins of PERV including Gag, Pol and Env,could not only were exist at the genome of minipig, but also been transcriped into RNA.The multiple infection of subtype A and subtype B was up to 85.1ï¼…in the GXBMminipig.Semi-quantitative RT-PCR was used to explore the PERV-mRNA distribution ofGXBM minipig in 9 tissues from tow pigs, Included liver,lung,pancreas,kidney,heart,spleen,thymus,ovary,peripheral blood and so on.The highest level ofPERV-mRNA was observed in the minipig kidney and peripheral blood and the leastwas in the pancreas. No significantly different were found among other tissues.Meanwhile, in order to understand PERV-env gene mutation in different generations in GXBM minipig. A total of 18 clones, which includied seven generationsand two groups kindred generations, were sequenced and were performed underPhylogenetic analyses. The 18 clones were grouped subtype A. Although some regularlysites mutations were found in different generations, no effect on deduced epitopes ofEnv. |
