In this study ,to produce a kind of vaccine including cross-protection in chickens, recombinant fowlpox virus (rFPV) expressing HA5 and co-expressing HA9 gene of AIV were construct. After construction of transfer vector,it was transfected on CEF cells with FPV F-017.Following clonning and purefication of blue plaque ,the pure HA-fowlpox virus recombinants were obtained,an did entified by restriction enzymean alysis and PCR.There sults show edthatthe recombinant fowlpox virus contained HA gene of AIV and had the stable genetic properties.There sult of Western-blot show edthatthe recombinant fowlpox virus expressed the hemagglutinin glycoprotein of AIV efficiently.Sixty-day–old SPF chickens were immunize and challenged three weeks later.After challenged with 50XLD, of HPAIV isolate per chicken, all chickens vaccinated with the recombinant fowlpox virus,the control chickens were died. The chickens vaccinated with recombinant fowlpox virus had HI antibody titer of 5.8 1og2 at 7 days post vaccination, and HI antibody titer rised to peak at 14 days post vaccination.HI an tibody was detactable at 14 days post vaccination.None Hl antibody were detected in the control or fowlpox virus immunized chickens before or after immunization nor challenge.H5 AIV Challenge virus was not reisolated from chickens vaccinated with the recombinant fowlpox virus,and detected in every one of all controld chickens.Virus was detected more frequently in long of control chickens.Determination of Tlymphocytes indicated that vaccination of recombinant fowlpox virus stimulated efectively cell-mediated immune response of chickens and activated TIymphocytes.The rFPV vaccine will be promising substitution of conventional whole virus inactivated vaccine to provent and control AIV.
