Influenza is an acute, hyperinfected disease caused by influenza virus and spreads broadly in human and animals. AIV is the short for Avian influenza virus, which is bird generic contract disease syndrome caused by influenza virus A. The high avian influenza virus (HAIV) has a strong pathogenicity, confines the use of avian influenza vaccine. At present, amantadine, rimantadine, zanamivir and oseltamivir carboxylate can be used for prophylaxis and treatment of avian influenza viruses. But they're have lower effects, higher adverse reactions and drug resistances. Before of advanced active vaccine, it is urgent to find new drugs that have high performance, low toxicities and independent intellectual property rights of China. Polyoxometallates possess distinct antiviral effects and low toxicities, become the hot spot of researching antiviral new drug increase. In this thesis, toxicities, anti-viral activities and immunoregulatory activities were measured in vitro and in vivo.1. Toxicities of PTW in vitro: Toxicities of polyoxometallates in vitro were measured by the methods of cultivation in cells. The results showed that TC50 in cells PTW is higher than AMA.2. Toxicities of PTW in vivo: Toxicities of PTW in vivo was measured by the methods of acute toxicity testing. The results showed that median lethal dose (LD50) of PTW is 2220 mg/kg. Hint: PTW is lowly toxic chemical materials.3. Anti-avian influenza virus activities of PTW in vitro: Anti-avian influenza virus activities of compounds in vitro were measured by the methods of cultivation in cells binding methyl thiazolyl tetrazolium(MTT). The results showed that TI is higher than AMA. Hint: PTW possess anti-avian influenza virus activities in vitro.4. Anti-avian influenza virus activities of PTW in vivo: Anti-avian influenza virus activities of PTW in vivo were measured in mice infected with avian influenza virus. The results showed that PTW depressed death rate, elevated survival rate and extended average living days of mice infected with avian influenza virus. PTW depressed lung humid weight, lung pathological degree, virus multiplication of lung and pathological change of mice infected with avian influenza virus significantly. PTW inhibited pneumonic potency of mice infected with avian influenza virus effectively. Hint: PTW possess anti-avian influenza virus activities in vivo.5. Immunoregulatory activities of PTW: Immunoregulatory activities of PTW were measured by the methods of organ weighter,T Lymphocyte transformation and NK Cytotoxicity activity study. The results showed that PTW stimulate spleen cells and thymocyte cells proliferation, promoted T Lymphocyte transformation function, NK cytotoxicities activity. Hint: PTW possesses immunoregulatory activities, elevated organism immunologic function, adjust antiviral activities.Conclusion: PTW possess lower toxicities in cells. PTW possess anti-avian influenza virus activities in vitro. PTW 25μg/mL dose possesses immunoregulatory activities.
