A strategy to enhance immune responses to conventional vaccines and DNA vaccines is very important in further development of the vaccine to control animal infectious diseases. The immunogenicity of vaccines can be significantly elevated with the use of powerful adjuvant.With the development of molecule biology and immunology,cytokines have been used as immunoasjuvants.Although cytokine can strongly trigger humoral and cellular immune responses to certain antigens or pathogens,main disadvantages of cytokines as adjuvant include high cost, difficult to produce and purify, short-lived bioactivity, and the need for frequent boosts.Interleukin-4(IL-4) is a versatile cytokine that plays an important role in the regulation of immune responses.It is engaged in regulating the biological activies, such as interleukin gene expression ,cell proliferation, B cell differentiation,T cell activation,and neutrophil and macrophage differentiation.Now few are known about the effect of mouse interleukin 4 on the immunity and the immune responses to vaccination.Furthermore,there is still lack of systemic explorations on the effect of transgenetic expression of mouse IL-4 gene in vitro and vivo.Especially,there is not reports about mouse interleukin-4 eukaryotic expression plasmid as the gene adjuvant transfection animals with somatostatin gene vaccine in internal and abroad.But mouse interleukin-4 react very important effect in regulation of immune defence and immune response,so we progressed mouse spleen cell interleukin-4 gene′s clone and transgenic expression in vivo,and investigate the effect of interleukin-4 gene expression to somatostatin gene vaccine′s result.Our experiment is expected to develop a novel immune potentiator to increase the immunity and enhance the immune responses of somatostatin gene vaccine.Otherwise,interleukin-6 is one important cytokine.It can enhancement multitude cells generation and differentiation.Respecting interleukin-6 important function is enhancement humoral and cell immunity, and there is not reports about mouse interleukin-6 eukaryotic expression plasmid as the gene adjuvant transfection animals with somatostatin gene vaccine in internal and abroad.Then mouse interleukin-6 gene expression plasmid may inhibit growth of neoplastic cell in mouse vivo, further prolong life period of bearing cancer′s mouse,and mouse interleukin-6 have predominance action to virus and generate strong protective immunity.First of all,we isolation mouse spleen cell.Mouse interleukin-4 and interleukin-6 cDNAs were synthesized by RT-PCR from mRNA from mouse spleen cell,which were stimulated by 5μL/mL Con A in vitro.The sequence of cloned mouse interleukin-4 and interleukin-6 cDNAs were determined.The length of the IL-4 is 423bp and the IL-6 is 623bp.The sequences of mouse IL-4 and IL-6 genes from mouse spleen cell were sunbmitted to GeneBank and registered as NM021283.1 and NM031168.By blasting the homologous sequences in GeneBank databases, the homology of interleukin-4 amino acids sequence from lymphocyte between NM021283.1 is 100%,and the homology of interleukin-6 amino acids sequence from lymphocyte between NM031168 is 99.69%.Consequences indicated that doing well gain the IL-4 and IL-6 genes in the experiment.The eukaryotic expression plasmid was constructed and named pIRES-IL-4M,pIRES-IL-6MThe eukaryotic expression plasmid were constructed and named pIRES-IL-4M,pIRES-IL-6M Investigating the expression of mouse IL-4 and IL-6 genes eukaryotic expression plasmid in mouse vivo.We approach mouse IL-4 and IL-6 ccommodation to somatostatin gene vaccine′s effect.Expected mouse IL-4 and IL-6 as gene adjuvant can sublimate antibody level of somatostatin,preferably balance out somatostatin in vivo,make growth hormone level corresp accrescence,to achieve the objective that better utility growth promotion.
