Toona sinensis belong to defoliation arbor with Ridgepole branch and excessive year living,which not only made as fasting timber,also as good vegetable with wood root,Its buds and shoots leaf spice thicking,flavor delicious.Furthermore,Toona sinensis have be provide with medicine-using to some extent.Toona sinensis extensively distributed to our country,resource was abundance,variety was rich relatively,due to many reasons make its bud-vegetable yield was very low,quality was not rather superiority.Additional,raise up seed shows to seriously separate in characters,taking root difficult in adopt transplant methods,many culturing seedling ways can not meet to need quantity in production and living.Especially,some superior and famous vegetable-using variety was slowly spreading and developing,thinks to effect on propagate in methods and speeds.In order to seek after a effective approach to improve Toona sinensis superior seeds quick-breeding technique,Tissue culture technology was introduced and studied profoundly in this paper.This method based on conservation female parent choiceness nature,simultanetity,it can quickly enlarge propagation in short period.Expect to explore a new way in this reseach-field.At present,there are less researches and reports in Toona sinensis tissue culture technique under parting-body condition.Moreover,its study results merely on primary experiment period,so there are many difficult to spreading apply to practice.This research base on plentiful known study information,studying and looking for tissue culture conditions of new variety-series T4 with have both traits of spice vegetable and superior timber which choosing and breeding by formerly our research staff group in west-north institute of botany.Look forward to suitable tech-method system was built,provide to tech-theory foundation on Toona sinensis quick-breeding.The research contents and results were introduced as followings:1.Toona sinensis optimem inoculating time is the middle ten-day of June~the last ten-day of July,this time is Toona sinensis speed-grow period.Half-lignifing bud segments may be fit to act as explants.2.The suiting culture medium for subculture and shoot multiplication was taken as MS, mineral reduce would block shoots well-balance growth.3.While GA3 and 6-BA concurrence in medium,the best effect on improve shoots growth and multiplication will be approach,the optimem multiplication medium was MS+0.2mg/L 6-BA+2.0mg/L GA3.4.Added PP333 or CCC at different concentrations to multiplying media when the shoots of Toona sinensis cultured.The basic medium for multiplication was MS+0.2mg/L 6-BA +2.0mg/L GA3,the results shows that PP333(10mg/L) or CCC(70mg/L) improved the qulitof shoots obviously,the effect of PP333(10mg/L) is better.5.The induce experiment of root results showed that single IBA was better,when IBA concentration was 1.0mg/L,rate of root was highest,it approach to 73%.Under condition of dark and illumination,the effect on root not too serious.But when the time of dark than 12d,it may be result shoots and leavies changed yellow and white.6.The proper root medium is based on single 1.0mg/L IBA in 1/2MS medium ( Saccharose 15g/L),append to 0.1mg/L PP333 and 10mg/L CCC,rooting and plantlets were strong,constrasted two kinds of hormons,experiment effect of 10mg/L CCC was evidently.7.Before transplant of plantlets,the effect of exercising plantlets was better based on seal in nature light 5-7d,then open seal exercising plant 3-5d.8.The fresh Vermiculite should be the most proper transplant medium materials.Sand could be take second place,river-sand act as cheapness and convenience medium,it also reach better transplant effect in certain condition.Additional, the suitable transplant time was from Mar to July annually,plantlets could keep high livability,quick grows and ease to manage in this period transplant.
