Channa maculate is a rare freshwater fish in China. In order to acquire further information about metabolic physiology of this breed, to accumulate basal parameter for histological and cytological studies and to establish principle on therapy of various diseases, the peripheral blood, liver and spleen were firstly selected as the materials of this work which was focused on histology and cytology. Secondly, The total RNA of liver was extracted and used as the template of cloning the uncoupling protein 2 (UCP2) gene which was regarded as one of the candidate genes for obesity. At last, the obtained segments were done on sequencing analyzing.At beginning, the peripheral blood sample was heparinized from the heart of fixed living fish and prepared as samples by traditional techniques, and then, using optical microscopy to observe and take photos. It appears that the erythrocytes have a high quantity and high density in peripheral blood. The shape of erythrocytes in maturation phase looks like ellipse. The nucleolus located in central part was containing compact chromatin mass and cytoplasm which were full of hemachrome. The erythrocytes in immaturation phase, which look like round or anomalistic and are easy to be distorted if they are pressed, can be found occasionally whose nucleolus is biggish including loose chromatin and whose cytoplasm filled with a small quantity of hemachrome. In addition, the leukocyte can be classified into groups such as thrombocyte, lymphocytes, monocyte and neutrophil with the absence of acidophil or basophil. The morphological characteristic of each group is basically similar to the observation of reported studies concerning teleostean blood cells. The thrombocyte, which is remarkably distinguished from other sort of cells, usually gathers in troops besides self-existent individual that figures small and elliptically, covered with discontinuous paper-thin cytoplasm going with rough surface or even uncovered in connection with compact chromatin.
