| With the combination of the technique of BIOLOG, PCR- DGGE and traditional plate count method, the black soil with fixed position appling fertilizer for twenty-six years were studied. The soil microbial quantity, activity, community structure and the metabolism function diversity were studied in the research. This provided the foundation to protect the classification authenticate of the soil microorganism and evaluate soil quality.The amount of the soil bacteria,actinomycete,fungus and a physiological microbe in the soil were meatured with traditional plate count method and most probable number MPN.The results showed that the proportion of bacterium, actinomycetes and fungi.in the microbial amount was 82.9%-98.9%,0.8%-17.1% and 0.1%-0.3% respectively.Applying fertilizer could increase the amount of microorganism while applying organic fertilizer could increase remarkably.organic fertility, fertilizer and organic fertility with fertilizer can increase the nitrogen nutrition most. And the rate of bacteria and actinomycetes in the soil is the highest with the tread of organic fertility with fertilizer. The organic material content is 29.4g/kg. The soil fertility of the treat organic fertility with fertilizer is the highest. So applying organic fertility with fertilizer can help to increase the fertility of soil.Because the amount of the bacterials shared the absolute advantage in the three most kinds of microorganisms in the soil. The amount of bacterials in the soil had the positive related relation (p<0.01) with organic material. The equation was y=54.302x-1184.9, Related coefficient r=0.8334**. With the higher OM content, the more amount of microorganisms were in the soil So the microorganism amount could evaluate the fertility of soil as an important index.The changing of amount of soil microbial biomass C was detected with Trichoromethane Fumigation-Extraction in the different grow periods of soybean.The amount in fruiting period was the highest then lower until mature period.In the same grow periods the amount of soil microbial biomass C was organic fertility with fertilizer > fertilizer> organic fertility.In the different grow periods of soybean, the dynamic changes of bacterials amount determined the... |
