Soil fungi, as one of the most important component parts in soil microbial community, have a large number of hypha that always grow to be networks. Fungi always plays a major as decomposer in acid environments. Soil fungi can both fix and mineralize soil nutrients and also play an active part in soil aggregate formation, which should be very important in paddy soil system. Ergosterol is the most abundant sterol held in the cell membranes of most filamentous fungi, certain microalgae, protozoa and yeasts. As soil ergosterol is easy to be decomposed, it always serves as an indicator of soil fungi biomass. Ergosterol content can be varied by fungi species and their relevant habitats; however, it is very important to construct an testing method with high efficiency and accuracy. Fertilization and soil water management have great effect on soil microbial habitat, and long-term application of different chemical fertilizer can change many soil properties, such as physical, chemical and biological properties, which will finally influence soil fertility and productivity. Wetting and drying process can improve gas diffusion and water permeability, as well as soil structure. In addition, it can also change the soil microbial community. The change of soil microbial habitat may bring about the transformation of soil microbial community. The general objectives of this study are as follows, firstly, to apply the general soil ergosterol measurement method to paddy soil. Secondly, to make out soil microbial community and biological activity change due to long-term chemical fertilization. At last, to understand the effects of soil water management on soil microbial community and biological activity.The soil samples were collected from the long-term fertilization experiment plots, which are located in the Institute of Red Soil Science, Jiangxi province. Nine fertilization treatments are as follows, CK(No fertilizer), N(nitrogen), P(phosphorus), K(Potassium), NP(nitrogen+ phosphorus), NK(nitrogen + Potassium), NPK(nitrogen + phosphorus + Potassium), 2NPK(double dose of NPK), NPKOM(NPK + organic manure).
