Uncoupling proteins are proton transporters in mitochondrial inner membrane. It plays an important role in thermogenesis, body weight regulation and energy metabolism.We use molecular technology include PCR-SSCP and PCR-RFLP to study the polymorphisms of UCP3 gene in Qinchuan Cattle, Luxi Cattle, Yanbian Cattle,Simmental,Nili-Ravi Buffalo and Murrah Buffalo.Analysis of correlation of the polymorphisms and biochemical index in serum which can provide a prefererce for resistance breeding of bovine.The results were as follows:1. The technology of PCR-SSCP was used to study the polymorphism of 2nd,4th,5th,6th exon and 5th intron of UCP3 gene. We found that there was polymorphism only in 5th intron.(1)The genotypes of UCP3-intron 5 in six bovine populations were analyzed.the result showed that the gene frequency and genotype frequency were at Hardy-Weinberg equilibrium(P>0.05).(2)The prominace genotype in Qinchuan Cattle, Luxi Cattle and Simmenta was BB type, which is AB type in Yanbian Cattle and in buffalo is CC type.The prominence allele is B, which is C allele in buffalo.(3)The value of the polymorphism information content (PIC) showed that the six populations were in moderate polymorphism. The polymorphism is abundance and the variation is big.(4)Sequence for UCP3 gene intron 5 demonstrated that there were two mutations at 3328 and 3374, i.e. G→A and C→T, respectively inYellow Cattle, which is A→G transition at 3056bp position in buffalo.(5)The correlation between genetic markers and biochemical index in serum were analyzed by using general linear model (GLM).The result showed that UCP3-intron5 can affect the content of kalium, sodium and calcium in serum in Nili-Ravi Buffalo, and the individuals with CC genotype were lower.It could affect content ofβ-globin in Luxi Cattle,and the individual with BB were higher than AA type.It could affectα1-globin in Murrah Buffalo,and AB type were higher than AA type.There was a significant differerce in albumin ,α2-globin and the ratio of albumin and globin between cattle and buffalo.2.
