Doubled haploid population with homozygous loci can be derived from spontaneous diploidization or artificial induction of haploid plants with an ant mitotic agent and be used to construct permanent segregation population. It can be directly applied both to breeding practice and to modern genetics analysis such as molecular marking and so on. The techniques of establishing DH population include two key processes,the obtaining of haploid plants and the doubling of chromosome. At present there are two major approaches for obtaining haploid plants,anther culture of F1 plants and eliminataion of chromosemes. Both showed the imperfections of low obtaining frequency and low diploidization efficiency of monoplioded plants.Focusing on the solution of the problems,this experiment detaildely studied the factors influencing the efficiency of anther culture such as sucrose concentration, hormone combination,genotypes and Cu2+ concentration,and those influencing the obtaining frequency of haploid plants such as pollination times of maize pollens,wheat genotypes,sucrose concentration in the processes of wheat haploid immature embryos culture,searched after the methods for safely oversummering and high diploidization efficiency of haploid wheat plants,and surveyed the resistance against powdery mildew and agronomic traits of DH population. The main results of the research are as follows:1.The increase of the cultural efficiency of wheat antherEffects of different concentration of sucrose and different combination of hormone and genotypes on cultural efficiency of wheat anthers in vitro were studied in the experiment. The results showed that the different sucrose concentrations significantly influenced the induction of wheat anther calli,and the rational concentration of sucrose supplemented in the induction medium was 100g/L. Further,different hormone combinations in differentiation medium had significant effects on redifferentiation frequency of wheat anther calli. The differentiation frequencies of plantlets of three genotypes were all at the highest (59.09%,48% and 57.14% respectively) when the hormones 1.5 mg /L KT + 0.5 mg /L IAA were added into W14,compared with the differentiation frequencies of plantlets in the W14 medium added with another two hormone combinations.
