| Fresh feces of Giant Panda, collected without contamination, was inoculated into the common broths and the anaerobic meat liver soups respectively and then cultured for 24~48h in 37℃.Things of the above cultured, after diluted to the same gradient and inclulated on solid cutured mediums quantitatively, were cultured for 24~48h under the condition with oxygen and without oxygen. According to the different colonies frorm the solid culture mediums, they were isolated and purified. G+ spore bacili,G+ having no spore bacilli,G+ coccobacteira and G- bacilli were firstly confirmed through microscopic observation and then the identified tests were carried out, including oxygen needing,M.R and V-P,catclase,gelatin liquefaction,KNO3 redution,indole,ferment of saccharide and mellows,sodium citrate,NaCl-phily and so on.Finally,12strains of bacteria were identified from the Giant Panda's feces. They were Lactorbacillus rhamosus,Lactobacillus brevis,Bacillus polymyxa,Enterococcus dispar,BifidoBacterium bifidum,Peptostreptococcus tetradius,Corynebacterium mycetoides,Lactobacillus panis,,Escherichia coli,Proteus Hauser,Atopobium Collins,Clostridium. Afterwards has carried on the toxicity test and the drugsensitive test,the toxicity test result demonstrated Peptostreptococcus tetradius,Lactorbacillus rhamosus,Lactobacillus brevis,Enterococcus dispar,BifidoBacterium bifidum does not have the pathogenicity to the mouse,but the Atopobium Collins and Bacillus polymyxa cause the mouse death.26 antibiotics were chosen to do drug sensitive test of eight bacteria strains. The results showed that Amikacin, being highly sensitive to E.coli and low sensitive to lactobacillia, was chosen. MIC and MBC about Amikacin affecting E.coli was 16ug/ml and 32ug/ml by tube duple dilute. When Amikacin is MIC, after Amikacin and E.coli co-culturing E.coli was low sensitive to Amikacin,sinomin compositea,Ofloxacin,cefotaxime,clindamycin and could bear to Penicillinum. When Amikacin is MBC,the results showed that the living curve was from droping to rising during Amikacin and E.coli co-culturing. |
