46 accessions of Chinese melon cultivars were collected in this study, which from different seven provinces and cities, including 8 accessions from Heilongjiang Province,4 accessions from Liaoning Province, 8 accessions from Jilin Province,three varieties from Beijing, 2varieties from Tianjing,2 varieties from Shandong Province and 1 variety from Shanxi Province,this study was to analyze the genetice distance and to draw the dendrograme of cluster tree analysis using the markers of Simple Sequence Repeat.And to breed 28 cross one varieties based on the SSR analysis of 46 varieties,searching the probability by using the genetic distance to forcase the heterosis of melon, giving the reference to collect the parents for utilization of heterosis of melon.Using SSR markers to analysis 46 accessions, resulted bands of forty-six primers were stability and abundant. Total 187 alleles, which 130 of them had polymorphism, revealed the average of 3.729 alleles per SSR marker, and the molecular size was between 250bp~1750bp. 46 Chinese melon cultivars were divided into nine groups using NTSYS program and methods of multidimensional scaling and cluster tree analysis. 22 characterization melon accessions from 9 different groups were selected to analysis again, they were divided into 4 groups, and some of them have the nearly genetic distance.Selected parents from 22 varieties, bred 28 cross-one, cultivated parents and F1 in greenhouse, the physiological and biological characters of parents and F1 which incluing weight of singal melon, thickness of pulp, hardness, sugar/acid, solvent and Vc content were measured, calculated the high heterosis and middle heterosis, found the relationships between genetic distance and heterosis. Result showed that, it preferred selecting parents from different groups to from same group for increasing the weight of singal melon and thickness of pulp. However, it was contrary to hardness, solvent sugar/acid and Vc conten. The relationships between genetic distance and heterosis of sugar/acid in entire were remarkable correlated in multinomial curve.
