Abstract/Summary:
Lepidopterid accounts for approximately 80% of agricultural and forest pests. Mulberry field pest Bombyx mandarina Moore belongs to Bombycidae, Lepidoptera, which comes from the same ancestor as economic insect Bombyx mori L.The insecticide resistance research of Bombyx mandarina and Bombyx mori surely will afford important theoretical evidence for Lepidopteral pests control. Many researches have indicated that CYP6 family is related with insecticide resistance. Bombyx mandarina M is a kind of wild field insect. It's difficult to ensure the homogeneity of Bombyx mandarina M and its continuous supply in a whole year. In order to solve this problem, artificial hatching and feeding of Bombyx mandarina M were firstly researched and one of its CYP6 family cDNA was then cloned and studied.1.In the present research, overwintering eggs of wild silkworm were collected from fields and conducted with instant acid treatment, acid treatment after chilling, and acid treatment before hatching in spring. The results showed that instant acid treatment on the overwintering eggs of Bombyx mandarina was proper with specific gravity of HCl acid 1.065-1.075, 46℃for 5 minutes; acid treatment after chilling was proper with the specific gravity of HCl acid 1.092, 47.8℃for 6 minutes; acid treatment on hibernated eggs after chilling for 80-100 days was proper with specific gravity of HCl acid 1.092, 47.8℃for 6 minutes, and there was no trend of higher hatching achieved by longer chilling days between 80-100 d; acid treatment on overwintering eggs of Bombyx mandarina before hatching in spring with specific gravity of HCl acid 1.092, 47.8℃for 6 minutes could obviously shorten hatching stage and increase hatching rate.
