| As the distribution centre of Hippophae, China contains rich genetic resource of sea buckthorn. In this study, the genetic diversity of H. rhamnoides subsp.yunnanensis and H. rhamnoides subsp. sinensis and interspecific relationship among subspecies were studied using ISSR and cpDNA, and moreover, the genetic relation of five sea buckthorn subspecies, including H. rhamnoides ssp. yunnanensis, H. rhamnoides ssp. sinensis, H. rhamnoides subsp. rhamnoides, H. rhamnoides subsp. mongolica and H. rhamnoidess subsp. turkestanica,were discussed. The aim of this study was to provide basic data for phylogeny and protection of genetic resource of Hippophae. The main results were as follows:(1) The study showed that there was different variation in morphology characters of H. rhamnoides subsp. yunnanensis. The condition heterogeneity of different population resulted that differ populations owned the same morphology character have some difference. In three populations, the range of cv in morphology character was 1.44%-18.73%. Most variation of morphology characters of yunnan xiaozhongdian population was bigger than Yunnan geza population and Xizang lizhi population.(2) According to cpDNA data, there was different clade in H. rhamnoides subsp. yunnanensis, the homology between the first cldogram and the second cldogram was 42%, and the homology between the third cldogram and former two cldogram was 32%. It implied that the origin of cpDNA of H. rhamnoides subsp. yunnanensis isn't single origin. Accordingly, a hypothesis was obtained in the cpDNA study: H. rhamnoides subsp. yunnanensis was hybird origin and was a nature hybrid. Its process of gene introgression was bidirectional or heteromallous. Additionally, as one origin of cpDNA of H. rhamnoides subsp. yunnanensis was entirely uniform H. rhamnoides subsp. sinensis, it indicated that H. rhamnoides subsp. sinensis may be one of parents of H. rhamnoides subsp. yunnanensis.(3) The homology of twenty samples of H. rhamnoides subsp. sinensis from different... |
