Companied the inland waters aggravately (such as lakes and rivers), disturbed the habitats and overexploited of source of economic freshwater mussels, which are deeply damaged the natural sources of some economic mussels, even extinct. In order to protect and asses the natural freshwater mussel sources of Dongting Lake, RAPD tech was used to assemble and classify the fifteen species from nine genera of wild economic freshwater mussels from south of Dongting Lake, we have found the RAPD special amplified primers and bands from DNA printing map, and we also have contrasted the differential tissue DNA of freshwater mussel extract effect, the main results are as follows:1. Improved CTAB method was used to extract the six tissues DNA, (such as sexual gland, zool, mantle, adductor muscle, gill, liver). We revealed that the sexual gland DNA extract effect is the best, tissue grind easy, water incubate time is short, the bands are single and clear. Then is the adductor muscle, the tissue grind is more difficult, water incubate time is longer, the least is liver, the bands are hazy. In a word, we can use the improved CTAB method extract genome DNA of freshwater mussel's different tissues, and the DNA can be used to progress the AFLP and RAPD experiment.2. 11 random primers were selected from 60, the rate of 18.2% and we have obtained 1775 clear, repeated and steady RAPD bands from the 11 primers amplify effect, average of 43.5 bands per-primer, and the bands length is between 50 bp and 1.5kb. These bands are revealed the RAPD marker maps of fifteen species from nine genera in wild economic freshwater mussel from south of Dongting Lake.3. In this experiment, 11 special RAPD amplified primers and 15 RAPD special bands were found from nine species, such as Anodonta arcaeformis , Cuneopsis pisciculus, Schistodesmus lampreyanus, Ptychorhychus pfisteri, Arconaia nceolata, Cuneopsis capitata, Lamprotula leai, Lamprotula zonata and Lanceolaria grayana, These special bands can be regard as the basis of genetic classify in the nine genera freshwater mussels.
