Abstract/Summary:
Plant diseases are threatening agriculture. To date, people mainly employ chemical pesticide, which tends to lead to environmental pollution and drug-resistance. On the other hand, traditional hybrid plant methods meet with greater and greater difficulties. So induction resistance is becoming a new hot spot in this area. Oligochitosan (OC) is an effective elicitor of plant resistance. But the induction mechanism still remains unclear. We obtained 96 bands from OC-treated tobacco through DDRT-PCR, one of which is MAPK (mitogen-activated protein kinase) and constructed a transgenic tobacco (Nicotiana tabacum var. samsun NN) cultivar that is transformed by the anti-sense MAPK.My purpose is to study the role that MAPK plays in the OC induced resistance by investigating some differences of recognized plant disease-related indexes between transgenic and wild type tobacco.I tested the F0 transgenic tobacco for the transcription of sense MAPK to find that it was totally suppressed by the anti-sense sequence, In the progeny F1, line T1 100% contained the anti-sense gene, which could be used for further experiments.I demonstrated the novel MAPK gene plays a role in N-gene mediated hypersensitive reaction of tobacco. Then, by the delay of necrosis appearance, I suppose that MAPK play an important role in TMV proliferation and the intracellular move. By the macroscopic TMV inhibition test, it is clear that MAPK is of great importance in plant resistance against TMV.PAL activity is increased by OC-induction via MAPK pathway, and PAL can synthesis a serial of secondly metabolic compounds to increase plant resistance. OC can increase the POD activity and lasts for some time via the MAPK gene. But the background of POD is low probably because of the negative influence of POD on cell growth.
