The Lycoris genus which was prominent in fancy and medinine, was of great commercial value. The chromosome in Lycoris genus that changed a lot and easy to detect, was good material in cytobilogy and phylogentic evolution. Prior to this work, there has been no report on localization of any gene via FISH in the Lycoris genus. In the present thesis, with the combination of karyotye analysis, we reported physical location of 5SrDNA and 45SrDNA genes via fluorescence in situ hybridization in L.awera and L.radiata from Jiangsu and anhui province; we tried to cloned 18SrDNA and 5SrDNA fragments form L.longiflora. The results were as follows:1, We have established the technic of fluorescence in situ hybridization in Lycoris genus and the technical foundation for the widespread availability of FISH in L. genus.2, The Karyotype analysis results showed: the karyotype, belonged to 4A type, was 2n=33=15t+18st in L.radiata; the karyotype, belonged to 3B type, was 2n=16=6m+10t in L.awera. They were conformed to the Robertson's change. We infered chromosomes of type I were acrocentric chromosomes that held short arm.3, The results of localication of 45SrDNA showed: In L.radiata, 6 hybridization sites were detected at the end of short arm of acrocentric chromosomes 3 and 5, the loci were close to the centromere. In L.awera, 11 hybridization sites were detected, 10 of which were at the end of short arm of all 10 acrocentric chromosomes. 1 site was detected near the centromere of metacentric chromosome, while the other homologous one had no hybridization signal. The results in L.awera demonstrated all 10 chromosome of type I were not telocentric but acrocentric chromosome. The numbers of signals in L.awera were nearly twice of the signals in L.radiata. Such divergence of 45SrDNA gene between the two materials demonstrated they were in far relationship, and might indicate centric fusion was the possible way of chromosome evolution.4, The results of localication of 5SrDNA showed: In L.awera, the 5SrDNA genes have been located on centromere, vicinity of centromere region and the end of long arm. Hybridization sites of 5SrDNA genes have been located on acrocentric chromosomes of L. radiata. 3 sites were located at the end of the long arm of chromosome 1; 3 sites were located in the vicinity of centromere of chromosome 6; while on chromosome 5, 2 weak signals in the vicinity of telocentric region did not existed on the other two homologous chromosome which showed 1 signal only.We presumed the 3 chromosomes originated from different parents, or...
